摘要
线粒体在维持细胞稳态中发挥关键作用,线粒体功能异常与多种疾病密切相关。线粒体外膜复合物在维持线粒体的功能中具有重要作用,其可选择性地识别和导入蛋白质,也可以运输其他分子,包括代谢物、无机离子和核酸。线粒体外膜易位酶40(TOM40)是线粒体外膜复合物的成员之一,是蛋白质导入线粒体内必不可少的传导通道。近年来TOM40在阿尔茨海默病、帕金森病与痴呆、非病理性认知老化以及各种心血管疾病和肿瘤中的作用已被广泛研究,基于TOM40为靶点治疗线粒体功能障碍成为研究热点。
Mitochondria play an important role in maintaining cell homeostasis,and the abnormal mitochondrial function is closely related to the pathogenesis of many diseases.Mitochondrial function mainly depends on translocase of outer mitochondrial member complex,which can selectively recognize and import proteins,and transport other molecules,including metabolites,inorganic ions and nucleic acids.Translocase of outer mitochondrial membrane(TOM)40 is one of the TOM complexes and an essential pathway for protein import into mitochondria.In recent years,the role of TOM40 in Alzheimer′s disease,Parkinson′s disease with dementia,non-pathological cognitive aging,cardiovascular diseases and tumor has been extensively studied.Treatment of mitochondrial dysfunction targeting TOM40 has become a research highlight.
