细颗粒物及冷刺激对气道上皮细胞黏蛋白5AC的刺激诱导机制研究

摘要 目的 分析细颗粒物及冷刺激对气道上皮细胞黏蛋白5AC的刺激诱导机制。方法 构建细颗粒物(PM2.5)及冷刺激的染毒细胞模型,刺激诱导气道黏液高分泌的发生,并借助酶联免疫吸附试验(ELISA)法、细胞免疫荧光法检测不同条件下MUC5AC蛋白的分泌水平表达。结果 ELISA法检测下PM2.5+5℃刺激组的MUC5AC蛋白(0.764±0.004)μg/ml的分泌水平分别高于PM2.5刺激组(0.422±0.003)μg/ml、5℃刺激组(0.325±0.003)μg/ml及对照组(0.215±0.004)μg/ml,差异有统计学意义(P<0.05);细胞免疫荧光法检测中,PM2.5+5℃刺激组(0.758±0.005)μg/ml的MUC5AC蛋白的分泌水平同样高于PM2.5刺激组(0.455±0.005)μg/ml、5℃刺激组(0.328±0.003)μg/ml及对照组(0.225±0.006)μg/ml,差异有统计学意义(P<0.05)。结论 寒冷环境对PM2.5致呼吸道损害效应的影响明显,此数据可为寒冷地区空气污染所致气道炎症的防控提供更好的理论依据。 Objective To analyze the stimulation-induced mechanism of fine particulate matter and cold stimulation on mucin 5AC(MUC5AC) in airway epithelial cells.Methods A cell model exposed to fine particulate matter(PM2.5) and cold stimulation was constructed to stimulate and induce the occurrence of hypersecretion of airway mu-cus,and the secretion level and expression of MUC5AC protein under different conditions were detected by ELISA and cellular immunofluorescence.Results The secretion level of MUC5AC protein [(0.764±0.004)μg/ml] in the PM2.5+5 ℃ stimulation group was higher than that in the PM2.5 stimulation group [(0.422±0.003)μg/ml],the 5 ℃stimulation group [(0.325±0.003)μg/ml] and the control group [(0.215±0.004)μg/ml],the difference was statistically significant(P<0.05).The results of cell immunofluorescence showed that the secretion level of MUC5AC protein in the PM2.5 +5 ℃ stimulation group [(0.758 ±0.005)μg/ml] was also higher than that in the PM2.5 stimulation group[(0.455±0.005)μg/ml],the 5 ℃ stimulation group [(0.328±0.003)μg/ml] and the control group[(0.225±0.006)μg/ml],the difference was statistically significant(P<0.05).Conclusion The cold environment has a significant impact on the damaging effect of PM2.5 on the respiratory tract,and this data can provide a better theoretical basis for the pre-vention and control of airway inflammation caused by air pollution in cold regions.
