丹参素异丙酯对D-半乳糖致衰老大鼠肾脏的保护作用 被引量:1

Protective effects of isopropyl-3-(3,4-dihydro-xyphenyl)-2-hydroxypropanoate on kidney in aging rats induced by D-galactose

摘要 目的:探讨丹参素异丙酯(IDHP)对D-半乳糖(D-gal)致衰老大鼠肾脏组织的保护作用。方法:40只Wistar大鼠分为空白对照组、衰老模型组、维生素E(VE,100.0 mg/kg)组、高剂量IDHP(21.0 mg/kg)组和低剂量IDHP(7.0 mg/kg)组5组,每组8只。后4组大鼠皮下注射25 g/L D-gal 2 mL/(kg·d)制作衰老大鼠模型,VE组、高剂量IDHP组和低剂量IDHP组大鼠皮下注射相应浓度的药物2 mL/kg,1次/d,连续8周。计算各组大鼠肾脏指数,HE染色观察肾脏组织细胞形态变化,检测血清肌酐、尿素水平,肾脏组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)活性及丙二醛(MDA)和过氧化脂质(LPO)水平。结果:与衰老模型组相比,VE组、高剂量IDHP组和低剂量IDHP组大鼠肾脏指数升高;与低剂量IDHP组相比,VE组、高剂量IDHP组大鼠肾脏指数升高(P<0.05)。衰老模型组大鼠肾脏组织呈现典型衰老性病理改变,VE组、高剂量IDHP组、低剂量IDHP组均有改善,但VE组、高剂量IDHP组改善更为明显。与衰老模型组相比,VE组、高剂量IDHP组和低剂量IDHP组大鼠血清肌酐、尿素水平降低;与低剂量IDHP组相比,VE组、高剂量IDHP组大鼠血清肌酐、尿素水平降低(P<0.05)。与衰老模型组相比,VE组、高剂量IDHP组和低剂量IDHP组大鼠肾脏组织中SOD、GSH-Px、CAT活性升高,MDA、LPO水平降低;与低剂量IDHP组相比,VE组、高剂量IDHP组大鼠肾脏组织中SOD、GSH-Px、CAT活性升高,MDA、LPO水平降低(P<0.05)。结论:IDHP具有延缓大鼠肾脏衰老的作用,这可能与其抗氧化作用有关。 Aim:To investigate the protective effects of isopropyl-3-(3,4-dihydro-xyphenyl)-2-hydroxypropanoate(IDHP)on the kidney in aging rats induced by D-galactose(D-gal).Methods:A total of 40 Wistar rats were randomly allocated into 5 groups,the blank control group,the aging model group,the vitamin E(VE,100.0 mg/kg)group,the high-dose IDHP(21.0 mg/kg)group and the low-dose IDHP(7.0 mg/kg)group,with 8 rats in each group.The aging rats were induced by subcutaneous injection of D-gal,meanwhile,the rats of the VE group,the high-dose IDHP group and the low-dose IDHP group were treated with daily subcutaneous injection of 2 mL/kg corresponding drugs for 8 weeks.In each group,the inferior vena cava blood of all the rats were used to detected the serum creatinine and urea levels;the kidney of rats in all the groups were taken out,the kidney index was calculated,half the kidney samples were used to observe the cell morphological changes of renal tissue by HE staining,while the other half were used to detect superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),catalase(CAT),malondialdehyde(MDA)and lipid peroxides(LPO).Results:Compared with the aging model group,the kidney index was higher in the VE group,the high-dose IDHP group and the low-dose IDHP group(P<0.05).Compared with the low-dose IDHP group,the kidney index was higher in the VE group and the high-dose IDHP group(P<0.05).The kidney tissue of the aging model group showed typical aging pathological changes,which was improved in the VE group,the high-dose IDHP group and the low-dose IDHP group,and especially in the VE group and the high-dose IDHP group.Compared with the aging model group,the serum creatinine and urea levels were lower in the VE group,the high-dose IDHP group and the low-dose IDHP group(P<0.05).Compared with the low-dose IDHP group,the serum creatinine and urea levels in the VE group and the high-dose IDHP group were lower(P<0.05).Compared with the aging model group,the SOD,GSH-Px and CAT activity were higher,and the MDA and LPO levels were lower in the VE group,the high-dose IDHP group and the low-dose IDHP group(P<0.05).Compared with the low-dose IDHP group,the SOD,GSH-Px and CAT activity were higher,and the MDA and LPO levels were lower in the VE group and the high-dose IDHP group(P<0.05).Conclusion:IDHP may delay the aging of kidney in rats,which may be related to its antioxidant effects.
