摘要
基于Amplex Red荧光分析法,构建新型乙酰胆碱酯酶抑制剂(acetylcholinesterase inhibitor,AChEI)高通量筛选模型并对其进行评价,检测97个灵芝(Ganoderma lucidum)子实体醇提物对乙酰胆碱酯酶(acetylcholinesterase,AChE)的抑制活性,再通过细胞模型进行验证。结果表明:构建的新型高通量AChEI筛选模型具有准确度、灵敏度高,不受测试样品溶液颜色干扰等优点;筛选出3个对乙酰胆碱酯酶具有较强抑制活性的灵芝子实体醇提物,研究结果可为后续灵芝来源AChEI的开发提供参考。
Based on Amplex Red fluorescence analysis,a novel high-throughput acetylcholinesterase inhibitor(AChEI)screening model was established and evaluated.Then the model was used to screen alcohol extracts of 97 Ganoderma lucidum fruiting body samples by measuring their inhibitory activity on AChE.The AcHE inhibition activity of samples selected by the model were verified by the SH-SY 5Y cell line.The results showed that this new high-throughput AChEI screening model had the advantages of high accuracy,high sensitivity and not being interfered by sample color.Using the model,three G.lucidum alcohol extracts were selected and thereby provided a reference for the development of AChEI from G.lucidum.
