摘要 【目的】以不同基因型百合花丝为外植体进行组织培养,研究培养基的适用性及花丝不同部位组织培养的难易程度。【方法】利用组织培养技术对花丝进行诱导扩繁。【结果】试验所用培养基适合不同基因型百合,‘白特里·昂菲特’和‘文雅王子’和‘白天堂’及亚洲百合新品系的优良单株(A1)花丝均能诱导分化出不定芽,对于大花卷丹花丝可诱导得到愈伤组织;属于不同基因型的材料其花丝组培难易程度存在差异,诱导分化率从高到低依次为‘白特里·昂菲特’和‘文雅王子’和‘白天堂’和亚洲百合新品系A1及大花卷丹;以花丝不同部位为外植体,诱导分化率存在差异,花丝基部>花丝中部>花丝顶部。【结论】培养基(MS+BA 1.0 mg/L+NAA 0.5 mg/L+糖50 g/L+琼脂5 g/L)适用于以不同基因型百合的花丝为外植体诱导愈伤或进一步形成不定芽,而且尤以花丝基部诱导分化率最高。该研究结果为在不损伤珍贵的优良单株和野生百合资源的前提下,通过花丝诱导进行组培扩繁,为进一步选育新品种和保存资源探索新途径。 【Objective】The filaments of different genotype lilies were used as explants for tissue culture to evaluate the suitability of medium and the difficulty in tissue culture using different parts of filaments.【Methods】The different parts of filaments were used as explants to be induced and propagated by tissue culture.【Results】The medium used in this study was suitable for different genotype lilies using which adventitious buds could be induced from the filaments of Lilium‘White Trionfitt’,Lilium‘Elegant Prince’,Lilium‘White Heaven’,and excellent individual plant of Asiatic hybrids(A1),and callus could be induced from the filaments of Lilium leichtlinii var.maximowiczii.There were differences in the difficulty of filament tissue culture among different hybrid lines.The order of induced differentiation rate from high to low were Lilium‘White Trionfitt’,Lilium‘Elegant Prince’,Lilium‘White Heaven’,A1,and Lilium leichtlinii var.maximowiczii.Using different parts of filaments as explants,the induced differentiation rate was also different(base>middle>top).【Conclusion】The medium(MS+Ba 1.0 mg/L+NAA 0.5 mg/L+sucrose 50 g/L+agar 5 g/L)was suitable for callus induction or further formation of adventitious buds using filaments of different genotype lilies,and the differentiation rate was the highest when the base of filaments was used as explants.This study provides a new way for tissue culture and propagation through filament induction without damaging the precious excellent single plant and wild lily resources,and for further new varieties breeding and resource conservation.
