摘要
目的 探讨辽宁地区幽门螺杆菌(H.pylori)阳性胃癌患者的癌组织中细胞空泡毒素基因A (vacA)优势基因型分布及其与肌肉增生蛋白3A (MEX3A)表达的关系。方法 选择2011年至2014年沈阳医学院附属中心医院外科手术切除的胃癌石蜡包埋病理标本98例,利用GT pureFFPE tissue DNA Extraction Kit提取DNA;利用PCR检测不同vacA基因型分布情况,用免疫组织化学染色法检测胃癌组织中MEX3A蛋白的表达情况,并分析二者的关系。结果 在H.pylori阳性胃癌中vacA s1、vacA m1、vacA s1m亚型分布高于其他vacA亚型(P <0.05),其中,vacA m1亚型与MEX3A表达密切相关(P <0.05)。MEX3A表达情况与肿瘤分化程度有关,差异有统计学意义(P <0.05)。结论 在辽宁地区H.pylori阳性胃癌患者的癌组织中,vacA m1为优势基因亚型,且表达与MEX3A有关,可能通过参与转录后调控相关细胞信号通路而影响胃癌发生、发展。
Objective To investigate the distribution of vaculating toxin geng A(vacA) dominant genotype and its effect on MEX3A protein expression in Helicobacter pylori(H.pylori)-positive gastric cancer tissues. Methods Totally 98 patients who underwent surgical excision of H.pylori-positive gastric cancer in the Central Hospital of Shenyang Medical College from 2011 to 2014 were enrolled in this study. DNA was extracted from the gastric cancer tissues using the GT pureFFPE tissue DNA Extraction Kit. vacA genotypes were determined through polymerase chain reaction,and MEX3A protein expression in the cancer tissues was detected using immunohistochemistry. Results The distribution of vacA s1,vacA m1,and vacA s1m1 subtypes was higher than that of other subtypes(P < 0.05).vacA m1 subtypes were closely related to the expression of MEX3A(P < 0.05),where MEX3A expression significantly correlated with the degree of tumor differentiation(P < 0.05). Conclusion In the Liaoning province of China,vacA m1 is the dominant subtype of H.pylori-positive gastric cancer. vacA m1 expression is related to MEX3A expression,and this may be involved in posttranscriptional regulation and related cell signaling pathways.
