影响细菌种群(太岁组织)结构关键因素的研究

Study on the Key Factors Affecting the Bacterial Population Structure of Taisui Tissue

摘要 探索了太岁取样部位和DNA提取前处理2个因素对细菌菌群结构的影响,并获知真实的优势菌群信息.对表里部位重复取样样品,施加水浸提、热溶解与微孔过滤组合成的3种前处理,提取的DNA用Miseq平台测序,挖掘出共有、占比高、有最高丰度OTU支撑的菌群.发现太岁中细菌种类丰富,大部分是杂菌,由表至内,多数类群不明原因消失,多样性和丰度降低,假单胞菌属(Pseudomonas)丰度显著升高;其与不动杆菌属(Acinetobacter)、根瘤菌属(Rhizobium)和鞘脂菌属(Sphingobium)为共有优势属;3种前处理方法获得的菌群结构近似,以内部组织直接热溶解过膜法为最优,其假单胞菌属占比达48.32%. In this researches,the effects of two factors were explored,the sampling site of Taisui and preDNA extraction on the structure of bacterial flora.The sometimes,the information on the dominant community of Taisui was obtained and the duplicate sample groups in different parts were extracted using different pretreatments by water immersion,thermal dissolution and micro-pore filtration.By determining the bacterium community structure,the Miseq platform analyzed and excavated the dominant bacteria with common,high proportion and maximum abundance supported by OTU.The results showed that there were many kinds of bacteria in Taisui and most of them are miscellaneous bacteria.Numerous bacteria disappeared for unknown reasons from outer to inner and diversity and abundance decreased.Moreover,the abundance of Pseudomonas increased significantly,and Pseudomonas was the common dominance with Acinetobacter、Rhizobium and Sphingobium.All the three pretreatment methods could obtain the same dominant group,however,the direct thermal dissolution of the internal tissue was optimal,by which Pseudomonas had a high proportion for 48.32%.
