M-CSF与TNF-α在子痫前期患者胎盘中的表达及意义分析 被引量:3

摘要 目的:探讨巨噬细胞集落刺激因子(M-CSF)与肿瘤坏死因子(TNF-α)在子痫前期(PE)胎盘中的表达及意义分析。方法:收集上海市长宁区妇幼保健院妇产科2019年8月到2021年8月50例PE患者的胎盘组织,根据类型不同分为早发型PE组(n=23)和晚发型PE组(n=27),另选取31例正常孕妇的胎盘组织为正常妊娠组。利用免疫组织化学SP法检测3组胎盘组织中M-CSF与TNF-α的表达情况,分析M-CSF、TNF-α与PE患者之间的关系。结果:早发型PE组和晚发型PE组的M-CSF平均灰度值、M-CSF阳性表达率均高于正常妊娠组(均P<0.05),且早发型PE组高于晚发型PE组(P<0.05);早发型PE组和晚发型PE组的TNF-a平均灰度值、TNF-α阳性表达率均低于正常妊娠组(均P<0.05),且晚发型PE组低于早发型PE组(P<0.05);相关性分析结果显示,M-CSF、TNF-α在PE胎盘组织中的表达呈负相关(r=-3.292,P<0.05)。结论:M-CSF、TNF-α与PE的病情轻重有关,可能参与了PE的发生发展。 Objective:To investigate the expression and significance of macrophage colony stimulating factor(M-CSF)and tumor necrosis factor alpha(TNF-α)in the placenta of women with preeclampsia(PE).Methods:The placental tissue samples were collect4ed from 50 PE women patients who stayed at Department of Obstetrics and Gynecology,Changning District Maternal and Children’s Healthcare Hospital of Shanghai between August 2019 and August 2021,then assigned to the early-onset PE group(n=23)and late-onset PE group(n=27)according to pattern of the disease onset.In addition,placental tissue samples were also collected from 31 women with normal pregnancy and assigned to the normal pregnancy group.The expression of M-CSF and TNF-αin placental tissues of the three groups were examined by immunohistochemical SP method.The relationship between M-CSF and TNF-αexpression in PE patients was analyzed.Results:The mean gray value of M-CSF and positive expression rate of M-CSF were higher in the early-onset PE group and the late-onset PE group than those in the normal pregnancy group(all P<0.05),and were higher in the early-onset PE group compared with the late-onset PE group(P<0.05).The mean gray value of TNF-a and positive expression rate of TNF-αwere lower in the early-onset PE group and the late-onset PE group than those in the normal pregnancy group(all P<0.05),and were lower in the late-onset PE group compared with the early-onset PE group(P<0.05).Correlation analysis showed that there was a negative correlation between M-CSF and TNF-αexpression in placenta tissue samples of the PE women(r=-3.292,P<0.05).Conclusion:M-CSF and TNF-αare associated with the disease severity,and may be involved in the development and progression,of PE.
