网果酸模提取工艺优化及其提取物对幽门结扎大鼠胃酸胃蛋白酶的影响 被引量:1

摘要 目的:确定网果酸模的最佳提取工艺并研究最佳提取工艺提取物对幽门结扎大鼠胃酸胃蛋白酶的影响。方法:采用L_(9)(3^(4))正交设计法,以网果酸模提取物中大黄素-8-β-D-吡喃葡萄糖苷、大黄酚-8-O-β-D-葡萄糖苷、酸模素、大黄素、大黄酚和大黄素甲醚6个成分总提取率及提取物抑菌环大小为考察指标,对网果酸模提取工艺进行优选。采用预防性给药原则,将48只SD大鼠随机均分为模型组、奥美拉唑组、奥美拉唑联合网果酸模组以及网果酸模高剂量组(0.780 g/kg)、中剂量组(0.390 g/kg)及低剂量组(0.185 g/kg),每组各8只,灌胃给药1周后采用幽门结扎建立胃溃疡大鼠模型,检测计算各组大鼠胃液总酸度、总酸排出量及胃蛋白酶活性。结果:网果酸模提取物抑菌环大小与大黄素-8-β-D-吡喃葡萄糖苷、大黄酚-8-O-β-D-葡萄糖苷、酸模素、大黄素、大黄酚和大黄素甲醚6个成分总提取率呈正相关。乙醇提取工艺最优,条件为6倍量75%乙醇,回流提取3次,每次90 min。网果酸模联合奥美拉唑可以极显著地抑制大鼠胃液释放(P<0.001),显著抑制胃酸排放(P<0.05)以及降低胃蛋白酶活力(P<0.01)并呈现一定剂量依赖性,网果酸模联合奥美拉唑给药对大鼠胃酸胃蛋白酶的抑制作用优于二者单独给药。结论:所确定的提取工艺稳定可行,网果酸模提取物具有抑制幽门螺杆菌和保护胃黏膜作用。 Objective:To determine the optimum extraction technology of Rumex chalepensis Mill.and study the effect of Rumex chalepensis Mill.extract on gastric acid and pepsin in rats with pylorus ligation.Methods:The total extraction rate of emodin-8-β-D-glucopyranoside,chrysophanol-8-O-β-D-glucoside,nepodin,emodin,chrysophanol and physcion,in Rumex chalepensis Mill.extract and the size of antibacterial ring of the extract were determined by orthogonal design.48 SD rats were randomly divided into model group,omeprazole group,omeprazole combined with Rumex chalepensis Mill.group and Rumex chalepensis Mill.High(0.780 g/kg),medium(0.390 g/kg)and low(0.185 g/kg)dose groups.One week after administration,gastric ulcer model was established by ligation of pylorus.The total acidity,total acid output and pepsin activity of gastric juice were detected and calculated.Results:The inhibition ring size of Rumex chalepensis Mill.extract was positively correlated with the total extraction rate of emodin-8-β-D-glucopyranoside,chrysophanol-8-O-β-D-glucoside,nepodin,emodin,chrysophanol and physcion.The optimum extraction conditions were as follows:6 times of 75%ethanol,refluxing for 3 times,90 min each time.The combination of the high-dose Rumex chalepensis Mill.and omeprazole significantly inhibited the release of gastric juice in rats(P<0.001),significantly inhibited gastric acid emission(P<0.05)as well as reduced pepsin viability(P<0.01),with statistically significant differences and showing some dose-dependent effects.The inhibitory effect of Rumex chalepensis Mill.combined with omeprazole on gastric acid and pepsin in rats was better than that of Rumex chalepensis Mill.and omeprazole alone.Conclusion:The extraction process was stable and feasible.Rumex chalepensis Mill.extract could inhibit helicobacter pylori and protect the gastric mucosa.
