摘要
目的:建立同位素内标稀释-超高效液相色谱串联质谱法测定花生及其制品中黄曲霉毒素B_(1)(Aflatoxin B_(1),AFB_(1))含量的方法。方法:用甲醇-水(70∶30,体积比)溶液进行提取,经冷冻离心,过滤,稀释后,加入同位素内标,waters C_(18)柱分离,正离子模式测定,内标法定量。结果:在0.01~100.00 ng·mL^(-1)内线性拟合度较高,3种加标水平的平均回收率为86.4%~102.1%,相对标准偏差小于10%,此方法检出限为0.01μg·kg^(-1),定量限为0.03μg·kg^(-1)。结论:该前处理方法简单快速,结果准确稳定,灵敏度高,可满足批量花生及其制品中AFB_(1)的检测需求。
Objective: To establish a method for the determination of AFB_(1) in peanut and its products by isotope internal standard dilution-UPLC-MS/MS. Method:Methanol-water(70∶30, volume ratio) solution was used for extraction.After freezing, centrifugation, filtration and dilution, isotope was added and separated by Waters C_(18) column, positive ion mode was determined and quantified by internal standard method. Result: The linear fit was high in the range of 0.01 ~ 100.00 ng·mL^(-1), the average recoveries of the three spiked levels were 86.4%~ 102.1%, and the relative standard deviation was less than 10%. The detection limit of this method was 0.01 μg·kg, and the limit of quantification was 0.03 μg·kg^(-1).conclusion: The previous method was simple and rapid, accurate and stable, with high sensitivity. It can meet the detection requirements of AFBin peanut and its productsin batch.
