DNA甲基化转移酶hsdM基因对维氏气单胞菌运动性的影响

摘要 为探究I型RM系统甲基转移酶基因hsdM对维氏气单胞菌运动性的影响,研究构建hsdM基因敲除的维氏气单胞菌突变株ΔhsdM,并探究该基因敲除对维氏气单胞菌生长、运动能力及鞭毛基因表达的影响。生长曲线测定结果表明:ΔhsdM菌株的生长速率在对数期生长后期开始缓慢降低,但总体生长水平与野生型基本一致;平板运动实验结果显示,hsdM基因敲除后,维氏气单胞菌运动性极显著下降;RT-qPCR结果表明,ΔhsdM菌株部分鞭毛基因表达显著下调。探究hsdM基因敲除对细菌表观性状的调节作用,为进一步探究N6-甲基腺嘌呤(6mA)甲基化调节基因表达的机制奠定理论基础。 To investigate the effects of methyltransferase gene hsdM of type I RM system on the motility of Aeromonas veronii,hsdM deletion strainsΔhsdM was constructed to explore the influences on the growth,motility and flagellum gene expressions of Aeromonas veronii.Growth curves showed that the growth rate ofΔhsdM strains began to decrease slowly in the late growth stage of logarithmic phase,while the overall growth levels of wild type andΔhsdM mutant are similar;motility assay suggested that the deletion of hsdM significantly reduced the motility of the bacteria.RT-qPCR indicated that the expressions of some flagellum genes were significantly down-regulated inΔhsdM strains.This study preliminarily explored the basic phenotypes of hsdM deletion related with bacterial motility,providing a theoretical foundation for further study on the mechanism of 6mA methylation on regulations of gene expressions.
