ADRM1对胰腺癌细胞增殖和转移的影响及相关机制研究

摘要 目的探讨黏附调节分子1(ADRM1)对胰腺癌细胞增殖和转移的影响及其相关机制。方法GEPIA数据库分析胰腺癌中的ADRM1 mRNA表达水平及其对患者预后的影响。EdU、CCK-8、Transwell实验分析抑制或过表达ADRM1对胰腺癌细胞增殖和转移的影响。采用生物信息学分析及Western blot探讨ADRM1调控胰腺癌细胞增殖和转移的相关机制。结果ADRM1 mRNA在胰腺癌中高表达,其表达水平与患者总生存率无关,与患者无病生存率有关,ADRM1 mRNA高水平组无病生存率低于ADRM1 mRNA低水平组。抑制ADRM1可以减弱胰腺癌细胞增殖和转移能力,过表达ADRM1可以增强胰腺癌细胞增殖和转移能力。ADRM1可以激活JAK酪氨酸蛋白激酶2-信号传导和转录激活因子3(JAK2-STAT3)信号通路。结论ADRM1作为促癌因子,在胰腺癌中高表达并促进胰腺癌细胞增殖和转移,其机制可能与激活JAK2-STAT3信号通路有关。 Objective To investigate the effect of adhesion regulating molecule 1(ADRM1)on the proliferation and metastasis of pancreatic adenocarcinoma cells and underlying mechanism.Methods GEPIA database was used to analyze the level of ADRM1 mRNA in pancreatic adenocarcinoma and its effect on clinical prognosis.The effects of silencing or overexpression of ADRM1 on the proliferation and metastasis of pancreatic cancer cells were analyzed by EdU,CCK-8 and Transwell assays.Bioinformation assay and Western blot were used to investigate the mechanism of ADRM1 in regulating the proliferation and metastasis of pancreatic cancer cells.Results The level of ADRM1 mRNA was up-regulated in pancreatic cancer,which was not correlated with overall survival,but was negatively correlated with disease-free survival.The high ADRM1 mRNA group showed lower disease-free survival than the low ADRM1 mRNA group.Inhibiting ADRM1 resulted in decreases in the proliferation and metastasis capacities of pancreatic cancer cells,while overexpression of ADRM1 enhanced the proliferation and metastasis of pancreatic cancer cells.ADRM1 activated the JAK tyrosine protein kinase 2-signal transduction and transcription activator 3(JAK2-STAT3)signaling pathway.Conclusions As an oncogene,ADRM1 is up-regulated in pancreatic adenocarcinoma and promotes the proliferation and metastasis of cells,which may be related to activation of the JAK2-STAT3 signaling pathway.
