人经血干细胞移植对宫腔粘连组织中α-SMA、TGF-β1及钙粘蛋白E含量的影响 被引量:5

Effects of human menstrual blood stem cell transplantation on the levels of α-SMA,TGF-β1 and E-cadherin in intrauterine adhesion tissues

摘要 目的研究人经血干细胞(MenSCs)接种到宫腔黏连组织表面,观察子宫内膜组织的结构变化,内膜组织中α-平滑肌肌动蛋白(α-SMA)、转化生长因子-β1(TGF-β1)及钙粘蛋白E含量变化,探索MenSCs接种改善宫腔黏连的机制。方法利用大鼠采用人为方法制造宫腔黏连模型,实验设计为对照组、模型组和MenSCs组,每组6只,共18只。比较3组内膜组织的结构改变和内膜组织中腺管的个数;Masson组织染色比较内膜组织的纤维化面积百分比;免疫印迹技术分析α-SMA、TGF-β1及钙粘蛋白E的蛋白条带的灰度值。结果与对照组比较,模型组内膜组织无上皮组织覆盖,只见结构增厚且无腺体的结缔组织,所含的纤维化面积[(0.70±0.05)%]较多(P<0.05),α-SMA(0.468±0.037)和TGF-β1(0.485±0.029)升高(P<0.05),钙粘蛋白E(0.242±0.021)降低(P<0.05)。与模型组比较,MenSCs组显示出现新生的上皮组织,腺体个数增加[(5.47±0.31)个],纤维化面积[(0.49±0.05)%]减少(P<0.05),α-SMA(0.370±0.025)和TGF-β1(0.387±0.016)降低(P<0.05),钙粘蛋白E(0.431±0.014)升高(P<0.05)。结论MenSCs接种到宫腔黏连组织表面后,能分化为子宫内膜上皮细胞,并能降低宫腔粘连组织中的α-SMA和TGF-β1蛋白含量,升高钙粘蛋白E的含量来改善宫腔黏连状况。 Objective To investigate the structural changes of endometrial tissue by seeding human blood stem cells(MenSCs)on the surface of intrauterine adhesions,and the content changes ofα-smooth muscle actin(α-SMA),transforming growth factor-β1(TGF-β1)and E-cadherin in the endometrial tissues,and to explore the mechanism of MenSCs inoculation in improving uterine adhesions.Methods The rat intrauterine adhesion models were establishe by factitiousness methods,then the experiment was divided into three groups:control group,model group and MenSCs group.The structural changes of the endometrial tissue and the number of glands in the endometrial tissue were observed and compared among the three groups.Moreover the degree of fibrosis was observed by Masson staining,and the gray value of protein bands ofα-SMA,TGF-β1 and E-cadherin was analyzed by Western Blot.Results Compared with those in control group,the endometrial tissue in model group was not covered by epithelial tissue,only thickened and without glandular tube connective tissue(P<0.05),with more fibrotic area(P<0.05),with higher levels ofα-SMA and TGF-β1(P<0.05),and lower values of E-cadherin(P<0.05).Compared with model group,MenSCs group showed new epithelial tissue and increased number of glandular tubes,decreased fibrosis area(P<0.05),decreased levels ofα-SMA and TGF-β1(P<0.05),and increased E-cadherin levels(P<0.05).Conclusion MenSCs can differentiate into endometrial epithelial cells after inoculated on the surface of uterine adhesions,which can decrease the contents ofα-SMA and TGF-β1 protein in intrauterine adhesions,and increase the content of E-cadherin to improve the intrauterine adhesion status.
