摘要
本实验旨在探讨双孢菇提取物对非酒精性高血脂大鼠血脂水平的影响及其作用机制。方法为将健康60只雄性SD大鼠随机分为6组:正常组,模型组,双孢菇高、中、低剂量组和阳性对照组,每组10只。除正常组喂食普通饲料外,其余各组采用高脂乳剂灌胃给药3周复制高脂血症动物模型。待模型建立后,正常组灌胃等量蒸馏水;高脂模型组继续灌胃高脂乳剂;药物组分别灌胃3.2 g/kg、6.4 g/kg、9.6 g/kg双孢菇提取物混悬液;阳性对照组灌胃9 mg/kg辛伐他汀。干预4周后,分别测定各组大鼠的体重、TG、CHOL等指标。取各组大鼠肝脏经10%福尔马林固定后,石蜡切片,HE染色行肝组织病理学观察。末次给药24 h后,大鼠尾静脉取血,采用酶标法检测各组动物血清ALT、AST、TG、TC、HDL、LDL等指标水平。结果:与模型组相比,双孢菇提取物高、中、低剂量组及阳性对照组大鼠的TG、CHOL、HDL-C、HCY,均低于模型组(P<0.05或P<0.01);双孢菇组和阳性对照组的ALT、AST水平均明显低于模型组(P<0.05或P<0.01)。组织切片结果显示双孢菇组的肝脏病变程度明显减轻。结论:双孢茹提取物能降低非酒精性高血脂大鼠TG、CHOL、HDL-C、HCY、ALT、AST等指标,能改善高脂血症大鼠肝损伤。
Objective:The purpose of this experiment was to investigate the mechanism of action and effect of Agaricus bisporus extract on non-alcoholic Hyperlipidemia rats.Methods:60 healthy male SD rats were selected and randomly divided into 6 groups.Normal group,model group,high-dose Agaricus bisporus extract group,medium-dose Agaricus bisporus extract group,low-dose Agaricus bisporus group and positive control group,extract10 rats in each group.The rates in the normal group were fed with ordinary diet and other five groups were gavaged with high fat emulsion in three weeks to copy hyperlipidemia animal model.After successful modeling,the normal group were gavaged isometric distilled water.the rats of hyperlipidemia model group were continued to be gavaged with high fat emulsion and drug groups were gavaged Agaricus bisporus extract suspension in 3.2g/kg,6.4g/kg,9.6g/kg separately.Positive control group was gavaged simvastatin in 9mg/kg.After 4 weeks of administration of irrigation,measuring the weight,TG,CHOL and other indexes of rats in each group.The liver sample from all rats were collected and fixed with formalin,and then conducted to make the paraffin section.Subsequently,HE staining were carried out to observe liver histopathology.24 hours after the last administration,collecting rat tail vein blood and using ELISA to test ALT,AST,TG,TC,HDL,LDL and other indexes in serum of animals in each group.Result:Compare with model group,the TG,CHOL,HDL-C and HCY of Agaricus bisporus extract high-dose,medium-dose,low-dose groups and positive control group were decreased in model group(P<0.05 or P<0.01);and the ALT,AST of Agaricus bisporus extract groups and positive control group were obviously decreased in model group(P<0.05 or P<0.01).Conclusion:Agaricus bisporus extract can decrease the TG,CHOL,HDL-C,HCY,ALT,AST and other indexes of non-alcoholic hyperlipidemia,and improve liver injury in hyperlipidemia rats.
