摘要
目的:观察肉蔻-5味丸对抑郁模型大鼠氨基酸类神经递质的影响,探讨其抗抑郁作用机制,为肉蔻-5味丸临床应用提供科学依据。方法:将60只SD雄性健康大鼠按体重随机分为正常组,模型组,肉蔻-5味丸高、中、低剂量组,氟西丁对照组6组,每组为10只大鼠。采用慢性轻度不可预知温和刺激结合孤立饲养方法建立抑郁模型。用体重增长情况、糖水消耗实验、Open-field敞箱实验等行为学方法评价抑郁模型成功与否。实验第一天开始造模同时给药,经28 d刺激后取血、海马组织,采用ELISA技术检测大鼠海马/血清谷氨酸(Glu)和γ-氨基丁酸(GABA)的水平。结果:与正常组比较,模型组体重增长缓慢,糖水消化量明显减少,Open-Field敞箱实验水平、垂直活动得分均明显降低,海马/血清Glu含量显著升高,GABA含量降低。与模型组比较,氟西丁对照组和蒙药组均有不同程度抑郁模型大鼠行为学改变,肉蔻-5味丸中、高剂量组具有明显的降低Glu水平、提高GABA水平。结论:蒙药肉蔻-5味丸对抑郁模型大鼠氨基酸类神经递质具有调节作用,调节抑郁模型大鼠氨基酸类神经递质含量可能是其抗抑郁作用机制之一。
Objective To observe the effect of Mongolian medicine Cardamom-5 Wei Wan on serum amino acid neurotransmitters in depression model rats,further explore the mechanism of antidepressant action,and provide scientific basis for its clinical application.Methods 60 healthy SD male rats were randomly divided into 6 groups according to their weight:normal group,model group,high dose group,medium dose group,low dose group and western medicine control group,with 10 rats in each group.Chronic mild unpredictable stimulation combined with isolated feeding was used to establish a depression model.The success or failure of the depression model was evaluated by behavioral methods such as sugar water consumption experiment and open-box experiment.On the first day of the experiment,the model was made and drugs were administered.After 28 days of stimulation,blood and hippocampal tissue were taken.The contents of amino acids such as Glu and GABA in hippocampus,serum were detected by ELISA.Methods:60 SD male healthy rats were randomly divided into six groups according to their body weight:blank control group,model group,Mongolian medicineCardamom-5 Weiwan high dose group,middle dose group,low dose group and Western medicine control group,with 10 rats in each group.The depression model was established by chronic mild unpredictable stimulation combined with isolated feeding.The success of depression model was evaluated by behavioral methods such as sugar water consumption experiment and open field open box experiment.After 28 days stimulation,blood and hippocampal 0tissue were taken.The contents of amino acid neurotransmitters such as Glu and GABA in hippocampus,serum were detected by ELISA.Results Compared with the normal group,the glucose digestion amount of the model group was significantly reduced,the horizontal and vertical activity scores of the open-box experiment were decreased,the hippocampal/serum Glu content was increased,and the GABA content was decreased.Compared with the model group,the western medicine group and the Mongolian medicine group both improved the behavioral changes of the depression model rats to varying degrees,while the middle and high dose Mongolian medicine groups significantly reduced the content of Glu and increased the content of GABA.Conclusion The Mongolian medicine Cardamom-5 Weiwan has a regulatory effect on amino acid neurotransmitters in depression model rats,and regulating the content of amino acid neurotransmitters in depression model rats may be one of its mechanisms of antidepressant action.
