摘要
目的探讨白细胞介素-1β(IL-1β)诱导甲状腺细胞子宫球蛋白相关蛋白1(UGRP1)的表达及其与Fas/FasL介导细胞凋亡的相关性。方法设立对照组、IL-1β组、IL-1β+抗FasL抗体组,体外培养大鼠甲状腺细胞(FRTL-5细胞),采用Real-time PCR法检测各组细胞UGRP1及Fas mRNA表达水平;采用流式细胞术检测各组细胞凋亡率。结果与对照组比较,IL-1β组、IL-1β+抗FasL抗体组UGRP1及Fas mRNA表达水平升高,差异有统计学意义(P<0.05);与对照组比较,IL-1β组甲状腺细胞早期凋亡率升高(7.49%±1.91%vs 28.46%±3.17%),差异有统计学意义(P<0.001),与IL-1β组比较,IL-1β+抗FasL抗体组甲状腺细胞早期凋亡率降低(28.46%±3.17%vs 19.20%±1.75%),差异有统计学意义(P<0.05);与IL-1β组比较,IL-1β+抗FasL抗体组UGRP1 mRNA(2.22±0.31 vs 2.66±0.28)及Fas mRNA(2.75±0.18 vs 3.03±0.16)表达水平差异无统计学意义(P>0.05)。结论IL-1β诱导甲状腺细胞UGRP1与Fas高表达以及细胞凋亡,UGRP1高表达与Fas/FasL介导的细胞凋亡不相关。
Objective To investigate the expression of uteroglobin-related protein 1(UGRP1) in thyroid cells induced by interleukin-1β(IL-1β) and its correlation with Fas/FasL mediated apoptosis. Methods Control group, IL-1β group and IL-1β+anti-FasL antibody group were established, rat thyroid cells(FRTL-5 cells) were cultured in vitro. The expression levels of UGRP1 and Fas mRNA of each group were detected by real-time PCR and the apoptosis rate of each group was detected by flow cytometry. Results Compared with control group, the mRNA expression levels of UGRP1 and Fas in IL-1β group and IL-1β+anti-FasL antibody group increased, and the difference was statistically significant(P<0.05). Compared with control group, the early apoptosis rate of thyroid cells in IL-1β group increased(7.49%±1.91% vs 28.46%±3.17%), and the difference was statistically significant(P<0.001). Compared with IL-1β group, the early apoptosis rate of thyroid cells in IL-1β+anti-FasL antibody group decreased(28.46%±3.17% vs 19.20%±1.75%), and the difference was statistically significant(P<0.05). Compared with IL-1β group, the expression levels of UGRP1 mRNA(2.22±0.31 vs 2.66±0.28) and Fas mRNA(2.75±0.18 vs 3.03±0.16) in IL-1β+anti-FasL antibody group were not significantly different(P>0.05). Conclusion IL-1β induces the high expression of UGRP1 and Fas and apoptosis of thyroid cells, the high expression of UGRP1 is not associated with Fas/FasL mediated apoptosis.
