左卡尼汀对慢性肾衰竭腹膜透析大鼠炎症因子、氧化应激及心脏保护作用影响

摘要 目的:探讨左卡尼汀对慢性肾衰竭腹膜透析大鼠炎症因子、氧化应激及心脏保护作用影响。方法:取30只大鼠(其中造模成功大鼠20只和正常大鼠10只)随机分为正常组、模型组和左卡尼汀组,各10只。实验组:给予左卡尼汀静脉注射,共4周;正常组和模型组静脉注射等剂量生理盐水,每日1次,共4周。大鼠慢性肾衰竭模型以5/6肾切除法建立,大鼠慢性肾衰竭腹膜透析模型以腹膜透析法建立。血清Cr和BUN水平以全自动生化分析仪检测;血清IL-6和TNF-α水平以酶联免疫吸附法测定;MDA水平以比色法测定,SOD活性以羟胺法测定;大鼠心肌细胞凋亡率以流式细胞仪测定。结果:模型组和左卡尼汀组大鼠血清Cr和BUN水平高于正常组(P<0.05);左卡尼汀组大鼠血清Cr和BUN水平低于模型组(P<0.05)。模型组和左卡尼汀组大鼠血清IL-6和TNF-α水平高于正常组(P<0.05);左卡尼汀组大鼠血清IL-6和TNF-α水平低于模型组(P<0.05)。模型组和左卡尼汀组大鼠血清MDA水平高于正常组,SOD水平低于正常组(P<0.05);左卡尼汀组大鼠血清MDA水平低于模型组,SOD水平高于模型组(P<0.05)。模型组和左卡尼汀组心肌细胞凋亡率高于正常组(P<0.05);左卡尼汀组心肌细胞凋亡率低于模型组(P<0.05)。结论:左卡尼汀可改善慢性肾衰竭大鼠肾功能,减轻炎症反应和氧化应激状态,且对心脏具有保护作用。 Objective:To investigate the effect of levonnitine on inflammatory factors,oxidative stress and cardioprotective effects in rats undergoing peritoneal dialysis with chronic renal failure.Methods:Thirty rats(including 20 successful model rats and 10 normal rats)were randomly divided into normal group,model group and L-carnitine group,with 10 rats in each group.In the experimental group,levocarnitine was injected intravenously for 4 weeks;The normal group and the model group were injected with the same dose of normal saline once a day for 4 weeks.The rat model of chronic renal failure was established by 5/6 nephrectomy,while the rat model of chronic renal failure was established by peritoneal dialysis.Serum Cr and BUN levels were detected by automatic biochemical analyzer;Serum IL-6 and TNF-αlevels were determined by ELISA;MDA level was determined by colorimetry and SOD activity by hydroxylamine method;The apoptosis rate of rat cardiomyocytes was determined by flow cytometry.Results:The levels of serum Cr and BUN in model group and levocarnitine group were higher than those in normal group(P<0.05);The levels of serum Cr and BUN in the levocarnitine group were lower than those in the model group(P<0.05).Serum IL-6 and TNF-αin model group and L-carnitine group.The level was higher than that of the normal group(P<0.05).Serum IL-6 and TNF-αin L-carnitine group.The level was lower than that of model group(P<0.05).The serum MDA level in the model group and the levocarnitine group was higher than that in the normal group,while the SOD level was lower than that in the normal group(P<0.05);The serum MDA level in the levocarnitine group was lower than that in the model group,while the SOD level was higher than that in the model group(P<0.05).Conclusion:L-carnitine can improve renal function,reduce inflammatory reaction and oxidative stress,and protect the heart in rats with chronic renal failure.
