正交优化黄连多糖提取工艺及其对α-葡萄糖苷酶的作用研究 被引量:4

摘要 目的 采用超声辅助法对黄连多糖提取工艺进行研究,并对其α-葡萄糖苷酶的作用进行研究。方法 在单因素研究基础上探究液料比、超声功率和提取时间对多糖提取率的影响。采用正交试验优化提取工艺,通过微量酶法检测黄连多糖对α-葡萄糖苷酶活性的作用。结果 黄连多糖提取的最佳工艺条件为液料比40 ml/g,提取功率100 W,提取时间30 min。采用优化提取条件进行工艺验证试验,测得多糖平均提取率为8.06%。提取的黄连多糖对α-葡萄糖苷酶活性具有一定的抑制作用,且其抑制类型呈典型的竞争性抑制。结论 超声辅助法提取黄连多糖工艺条件操作方便,重复性好,且黄连多糖具有α-葡萄糖苷酶的抑制作用,有开发为降糖药物的应用前景和市场潜力。 Objective To study ultrasound-assisted extraction of polysaccharides from Rhizoma Coptidis and its effect on α-glucosidase.Methods Based on the single factor study, the effects of liquid-to-liquid ratio, ultrasonic power and extraction time on the extraction rate of polysaccharides were investigated. The extraction process was optimized by orthogonal test, and the effect of flavonoids on α-glucosidase activity was detected by microenzyme method.Results The optimum process conditions for the extraction of polysaccharides from Coptis chinensis were liquid-liquid ratio of 40 ml/g, extraction power of 100 W, and extraction time of 30 min. The process validation test was carried out under optimized extraction conditions, and the average extraction rate of polysaccharide was 8. 06%. The extracted Coptidis Rhizoma polysaccharide has a certain inhibitory effect on α-glucosidase activity, and its inhibition type is typical competitive inhibition.Conclusion Ultrasound-assisted extraction of Coptidis Rhizoma polysaccharides is convenient and reproducible, and Coptidis Rhizoma polysaccharides have α-glucosidase inhibitory effect, and have potential applications and market potential for the development of hypoglycemic drugs.
