摘要
目的分析Bestrophin 2(BEST2)在结肠癌中的表达及其与结肠癌患者临床预后的关系,探讨BEST2对结肠癌细胞增殖的影响和作用机制。方法结合TCGA数据库和实时荧光定量PCR,分析结肠癌中BEST2的表达情况。在HT29细胞系中过表达BEST2,通过CCK-8和EdU实验探讨BEST2对结肠癌增殖的影响,使用流式细胞仪检测细胞内的活性氧(ROS)水平。结果BEST2在结肠癌组织及细胞中低表达,且BEST2低表达的结肠癌患者预后差。体外实验表明过表达BEST2抑制HT29细胞增殖。过表达BEST2使细胞内ROS水平升高,从而抑制细胞增殖。结论BEST2在结肠癌中低表达且其表达水平与结肠癌预后相关。BEST2通过调节细胞内ROS水平抑制结肠癌细胞增殖。
Objective To investigate the expression of Bestrophin 2(BEST2)in colon cancer(CC)tissues and cell lines and the relationship with clinical prognosis as well as its effect on the progression of CC cells and the potential mechanisms.Methods The TCGA database and the quantitative real-time polymerase chain reaction(PCR)were to analyze the expression level of BEST2 in CC tissues and cells.HT29 cell line was used in this study,and the cell function was explored after BEST2 overexpression.The effects of BEST2 on the CC proliferation were determined with CCK-8 and EdU assay.Intracellular reactive oxygen species(ROS)were detected by flow cytometry.Results The expression level of BEST2 in CC tissues and cells was significantly low.The overall survival of CC patients with low expression of BEST2 was lower than those with high expression of BEST2.Mechanistic studies suggest that BEST2 inhibits CC proliferation by promoting intracellular ROS levels.Conclusion The expression of BEST2 was decreased in CC specimens.Meanwhile,BEST2 inhibited the proliferation of colon cancer cells through a mechanism that involves the modulation of intracellular ROS levels.
