冠状动脉粥样硬化性心脏病患者血清中miR-7-5p和PARP1水平与心功能的相关性研究 被引量:4

Correlation between serum miR-7-5p and PARP1 levels and cardiac function in patients with coronary atherosclerotic heart disease

摘要 目的探讨冠状动脉粥样硬化性心脏病(CHD)患者血清中miR-7-5p和聚腺苷二磷酸核糖聚合酶1(PARP1)水平及其与心功能的相关性。方法前瞻性选取2020年5月至2021年5月上海交通大学医学院附属第九人民医院黄浦分院心脏科住院并经冠状动脉造影证实为CHD患者85例为CHD组。同时选取同期同院健康体检者85名为对照组。对两组研究对象的一般资料、血清中miR-7-5p和PARP1表达、心功能指标进行对比分析,并对不同心功能分级患者血清中miR-7-5p、PARP1水平及心功能指标进行对比,并分析miR-7-5p、PARP1与心功能指标的相关性。结果CHD组体重指数、收缩压、空腹血糖、低密度脂蛋白胆固醇均高于对照组,高密度脂蛋白胆固醇低于对照组,差异均有统计学意义(P<0.05)。CHD组血清中miR-7-5p的相对表达量为0.37±0.06,明显低于对照组(1.01±0.12),血清中PARP1的相对表达量为1.84±0.25,显著高于对照组(0.63±0.06),差异均有统计学意义(P<0.05)。CHD组中LVESD、LVEDD为(45.68±4.23)、(72.19±8.72)mm,明显高于对照组[(31.72±2.69)、(47.36±5.74)mm],LVEF、CO为(46.37±6.81)%、(5.53±1.69)mL/min显著低于对照组[(55.19±7.24)%、(7.31±1.78)mL/min],差异均有统计学意义(P<0.05)。Ⅳ级患者血清中miR-7-5p相对表达量、LVEF、CO均显著低于Ⅱ级和Ⅲ级,Ⅳ级患者血清中PARP1相对表达量、LVESD、LVEDD显著高于Ⅱ级和Ⅲ级,差异均有统计学意义(P<0.05)。CHD组血清中miR-7-5p的相对表达量与LVESD和LVEDD呈负相关,与LVEF和CO呈现正相关(P<0.05);CHD组血清中PARP1的相对表达量与LVESD和LVEDD呈正相关,与LVEF和CO呈负相关(P<0.05)。结论冠状动脉粥样硬化性心脏病患者血清中miR-7-5p的表达降低,PARP1表达升高,并且与心功能的指标具有一定的相关性。 Objective Investigate the correlation between serum levels of miR-7-5p and polyADP-ribose polymerase 1(PARP1)and cardiac function in patients with coronary atherosclerotic heart disease.Methods A total of 85 CHD patients admitted to the cardiology department of Huangpu branch of the Ninth People's Hospital Affiliated to the Medical College of Shanghai Jiaotong University from May 2020 to may 2021 and confirmed by coronary angiography were prospectively selected as CHD group,85 healthy subjects who were admitted to the hospital for physical examination during the same period were selected as the control group.General information of the two groups of subjects,the expression of miR-7-5p and PARP1 in serum,and the heart functional indicators were compared and analyzed,and the serum levels of miR-7-5p,PARP1 and cardiac function indicators in patients with different cardiac function classes were compared,and the correlation between miR-7-5p,PARP1 and cardiac function indicators was analyzed.Results The body mass index,systolic blood pressure and fasting blood glucose and low-density lipoprotein cholesterol of the CHD group were higher than those of the control group,and the high-density lipoprotein cholesterol was lower than that of the control group,and the differences were statistically significant(P<0.05).The relative expression of miR-7-5p in the serum of the CHD group was 0.37±0.06,which was lower than that of the control group(1.01±0.12),the relative expression of PARP1 in the serum of the CHD group was 1.84±0.25,which was higher than that of the control group(0.63±0.06),the differences were statistically significant(P<0.05).The LVESD and LVEDD in the CHD group were(45.68±4.23),(72.19±8.72)mm,which were significantly higher than the control group[(31.72±2.69),(47.36±5.74)mm],and the CHD group LVEF and CO were(46.37±6.81)%,(5.53±1.69)mL/min,which were significantly lower than the control group[(55.19±7.24)%,(7.31±1.78)mL/min],the differences were statistically significant(P<0.05).The relative expression of miR-7-5p in the serum,LVEF and CO of patients with gradeⅣwas significantly lower than that of gradeⅡandⅢ,the relative expression of PARP1 in the serum,LVESD and LVEDD of patients with gradeⅣwere significantly higher than gradeⅡandⅢ,the differences were statistically significant(P<0.05).The relative expression of miR-7-5p in serum of CHD group was negatively correlated with LVESD and LVEDD,and positively correlated with LVEF and CO(P<0.05).The relative expression of PARP1 in serum of CHD group was correlated with LVESD and LVEDD.LVEDD is positively correlated,and negatively correlated with LVEF and CO(P<0.05).Conclusion The expression of miR-7-5p in the serum of patients with coronary atherosclerotic heart disease is decreased,and the expression of PARP1 is increased,and it is related to the indicators of heart function Have a certain relevance.
