摘要 抗结核药物性肝损伤(ATB-DILI)发生率较高,加重结核病患者的诊疗负担,其发病机制是近年来的研究热点。炎症损伤机制是目前ATB-DILI研究最广泛的,涉及线粒体损伤和炎症因子分泌增加,线粒体损伤可导致基质金属蛋白酶(MMPs)表达升高。MMPs是一类可降解或重塑细胞外基质的肽链内切酶,其与ATB-DILI的发生关系密切。目前研究多集中于MMP-2、MMP-9和MMP-14,在肝损伤方面仍有众多MMPs的作用有待阐明。本文就MMPs的结构和分类、活化和调节以及其在肝损伤机制中的研究进展进行综述,为今后MMPs与ATB-DILI的相关性研究提供理论基础。 The pathogenesis of anti-tuberculosis drug-induced liver injury(ATB-DILI)has been a study focus in recent years as the incidence of which is high and increases the burden of treatment for patients.The mechanism of inflammatory injury is the most widely studied in ATB-DILI,involving mitochondrial injury and increased secretion of inflammatory factors,and mitochondrial injury can lead to elevated expression of matrix metalloproteinases(MMPs).MMPs is a class of endopeptidase that can degrade or remodel the extracellular matrix,which is closely related to the occurrence of ATB-DILI.Currently,most studies have focused on MMP-2,MMP-9 and MMP-14,but there are still many roles of MMPs in liver injury that remain to be elucidated.Accordingly,this paper reviews the study advances in the structure and classification of MMPs,and their activation and regulation mechanisms and mechanism of action in liver injury,aiming to provide a theoretical basis for future studies on the correlation between MMPs and AT B-DILI.
