摘要
目的:基于数据挖掘及网络药理学分析含枇杷叶核心药物组治疗咳嗽的作用机制。方法:以枇杷叶和咳嗽为关键词进行处方筛选,并进行处方标准化处理;对枇杷叶方剂实行关联规则分析,筛选枇杷叶核心药物组。在中药系统药理学数据库与分析平台收集枇杷叶核心药物组的成分及咳嗽相关的靶点。运用String 11.0构建交集靶点蛋白质-蛋白质相互作用网络,通过拓扑分析获得核心靶点,并进行基因本体论(GO)功能富集分析和京都基因与基因组百科全书(KEGG)富集分析。结果:数据库筛选获得治疗咳嗽的含枇杷叶标准方剂有69首,涉及136味中药,其中枇杷叶、川贝母、苦杏仁及甘草具有强关联性,以这4味中药组成的核心药物组共涉及71个成分及65个交集靶点,17个核心靶点有白细胞介素(Interleukin,IL)-6、信号转导和转录活化因子3(Signal Transducer and Activator of Transcription 3,STAT3)、IL-1β等;GO富集分析表明主要涉及对脂多糖及细菌来源分子的反应等生物学过程,KEGG信号通路分析表明主要与肿瘤坏死因子(Tumor Necrosis Factor,TNF)及IL-17等信号通路密切相关。结论:含枇杷叶核心药物组能通过多成分、多靶点、多通路的方式,从抗炎、抗病毒、抑菌方面发挥其治疗咳嗽的作用。
Objective:Based on data mining and network pharmacological analysis,the action mechanism of core prescriptions containing Pipa Ye(Eriobotryae Folium)in treating cough was analyzed.Methods:Pipa Ye and cough were used as key words for prescription screening and prescription standardization.The association rules of prescriptions containing Pipa Ye were analyzed to screen the core couplet medicines of Pipa Ye.The components of core couplet medicines of Pipa Ye and cough related targets were collected in TCMSP database.String 11.0 was used to construct protein-protein interaction network of the intersection targets.The core targets were obtained by topological analysis.The GO and KEGG enrichment analysis was performed.Results:There were 69 standard prescriptions containing Pipa Ye for cough in the databases,involving 136 kinds of Chinese materia medica,among which Pipa Ye,Chuanbeimu(Fritillaria Cirrhosa),Kuxingren(Bitter Almond)and Gancao(Licorice)had strong correlation.A total of 71 components and 65intersection targets were involved in the core couplet medicines composed of these 4 kinds of Chinese materia medica,and the 17 core targets included IL-6,STAT3,IL-1β and so on.The Go enrichment analysis showed that it was mainly involved in biological processes such as reaction to lipopolysaccharide and bacterial derived molecules.The KEGG signal pathway analysis showed that it was mainly closely related to TNF,IL-17 and other signal pathways.Conclusion:The core couplet medicines containing Pipa Ye can play its role in antiinflammation,anti-virus and anti-bacteria through multi-component,multi-target and multi-pathway.
