摘要
目的本研究拟评估没食子提取物对人牙周膜成纤维细胞(human periodontal ligament fibroblasts,HPDLF)的细胞毒性和储存后的增殖和凋亡状态,以期预防再植后的并发症。方法分离培养HPDLF,暴露于没食子等实验溶液,测量细胞活力,细胞增殖水平和细胞凋亡水平。结果当没食子浓度低于400 mg/ml时,未出现显著的细胞毒性(P>0.05);没食子提取物与实验组比较,HPDLF的细胞增殖显著增加,凋亡细胞减少(P<0.05),与细胞培养基组比较细胞增殖差异无统计学意义(P>0.05),而细胞凋亡水平显著高于培养基组(P<0.05)。结论没食子提高了HPDLF活力与增殖,降低了细胞凋亡水平,其可以成为有益于脱位牙储存的介质,提高其生理健康水平。
Objective To determine the cytotoxicity of gall extract on human periodontal ligament fibroblasts and its ability of proliferation and apoptosis after storage in order to prevent complications after replantation.Methods HPDLF was isolated,cultured and exposed to gall and other experimental solutions.The cell viability was detected by azurol reduction assay,the proliferation level of cells was measured by XXT assay,and the apoptosis level of cells was determined byflow cytometry.Results There was no significant cytotoxicity(P>0.05)when the concentration of gall was lower than 400 mg/ml.Compared with the experimental group,the cell proliferation of HPDLF was increased and the apoptotic cells were decreased significantly(P<0.05).Compared with the cell culture medium group,there was no significant difference in cell proliferation(P>0.05),but the level of cell apoptosis was significantly higher than cell culture medium group(P<0.05).Conclusion Gall can improve theactivity and proliferation of hPDLF and reduce the level of apoptosis.It can become a medium that is conductive to the storage ofdislocated teeth and improve their physiological health.
