摘要
目的:探讨刺五加改善持续光照条件下大鼠睡眠-觉醒节律的作用及脑内GABA、L-Glu含量变化。方法:利用CLAMS,以睡眠-觉醒节律和睡眠时间为观察指标,研究给药后刺五加改善持续光照条件下大鼠睡眠-觉醒节律的作用。利用ELISA法,研究给药后刺五加对持续光照条件下大鼠脑内GABA、L-Glu含量变化的影响。结果:正常光照组大鼠的睡眠-觉醒呈现昼夜节律变化,节律周期稳定。与正常光照组比较,9∶00-11∶00持续光照组睡眠时间减少;与持续光照组比较,9∶00-11∶00给药组睡眠时间增多;与正常光照组比较,21∶00-23∶00持续光照组睡眠时间减少,差异有统计学意义(P<0.05)。在11∶00与正常光照组比较,持续光照组GABA含量下降,L-Glu含量升高;与持续光照组比较,给药组GABA含量升高,差异有统计学意义(P<0.05)。在23∶00与正常光照组比较,持续光照组GABA含量升高,L-Glu含量下降;与持续光照组比较,给药组GABA含量下降,差异有统计学意义(P<0.05)。结论:刺五加能通过调节大鼠的节律分布改善大鼠的睡眠,其作用机制可能与影响L-Glu、GABA的含量变化,进而改善大鼠的睡眠-觉醒节律有关。
Objective:To investigate the effect of Acanthopanax senticosus on sleep-wake rhythm and the contents of GABA and L-Glu in the brain of the rats under continuous illumination.Methods:Using the experimental animal monitoring system CLAMS,taking sleep-wake rhythm and sleep time as observation indicators,the effect of Acanthopanax senticosus on improving sleep-wake rhythm in rats under continuous light conditions was observed.ELISA method was used to study the effect of Acanthopanax senticosus on the changes of GABA and L-Glu in the brain of the rats under continuous illumination.Results:In terms of sleep-wake rhythm,the sleep-wake of the rats in the normal light group showed circadian rhythm changes,and the rhythm cycle was stable.Compared with that in the normal light group,the sleep time of 9:00-11:00 was reduced in the continuous illumination group(P<0.05);compared with that in the continuous illumination group,the sleep time of 9:00-11:00 was increased in the medication group(P<0.05).Compared with that in the normal light group,the sleep time of 21:00-23:00 was decreased in the continuous illumination group(P<0.05).Compared with that in the normal light group at 11:00,the content of GABA was decreased,and the content of L-Glu was increased in the continuous illumination group(P<0.05);compared with that in the continuous illumination group,the content of GABA was increased in the medication group(P<0.05).Compared with that in the normal light group at 23:00,the content of GABA was increased,and the content of L-Glu was decreased in the continuous illumination group(P<0.05);compared with that in the continuous light group,the content of GABA was decreased in the medication group(P<0.05).Conclusion:Acanthopanax senticosus can improve the sleep of the rats by regulating the rhythm distribution,the mechanism may be related to the changes of GABA and L-Glu,and then improve the sleep-wake rhythm of the rats.
