固本防哮饮对哮喘合并食物过敏小鼠NLRP3炎症小体通路的影响

摘要 目的:基于NLRP3炎症小体通路探讨固本防哮饮防治哮喘合并食物过敏小鼠气道炎症的作用机制。方法:BALB/c小鼠采用卵蛋白(OVA)灌胃和滴鼻致敏,建立气道炎症模型和哮喘合并食物过敏的联合动物模型,随机分为正常组(control)、气道模型组(AR)、联合模型组(FAR)、固本防哮饮组(GBFXD)及孟鲁司特钠组(Mont)。模型组采用固本防哮饮和孟鲁司特纳灌胃治疗,末次灌胃处死,取肺组织进行HE染色,免疫荧光法检测肺组织中连接蛋白claudin-1、occludin、E-cadherin的表达,Western blot法检测肺组织中caspase-1、NLRP3、NF-κB、IL-1β的表达。结果:联合模型组的炎症甚于单纯哮喘气道模型组(P<0.05),与联合模型组相比,固本防哮饮组的肺部炎症得到改善(P<0.05)。与联合模型组相比,固本防哮饮组的肺组织中occludin-1、E-cadherin显著增加(P<0.05)。与联合模型组相比,固本防哮饮组的肺组织中caspase-1、NLRP3、NF-κB、IL-1β表达水平显著下降(P<0.05)。结论:固本防哮饮可能通过调控NLRP3炎症小体通路,保护气道上皮屏障的完整,进而减轻哮喘合并食物过敏后的气道炎症反应。 Objective:To investigate the mechanism of Guben Fangxiao Decoction(GBFXD)in preventing and treating respiratory inflammation of asthma accompanied with food allergy based on NLRP3 inflammasome pathway.Methods:BALB/c mice were sensitized by ovalbumin(OVA)gavage and nasal drops to establish the model of respiratory inflammation of asthma accompanied with food allergy.The successful modeling mice were randomly divided into the respiratory allergy(AR)group,the food and respiratory allergy(FAR)group,the GBFXD group and the Montelukast Sodium(Mout)group;the normal BALB/c mice were taken as the blank control.24 hours after the last administration,the mice were sacrificed and the lung tissues were taken for HE staining.The protein expressions of claudin-1,occludin and E-cadherin in lung tissues were detected by immunofluorescence method.The expressions of caspase-1,NLRP3,NF-κB and IL-1βin lung tissues were detected by Western blot.Results:The inflammation in the FAR group was more severe than that in the AR group(P<0.05),and the lung inflammation in the GBFXD group was improved compared with that in the FAR group(P<0.05).Compared with those in the FAR group,the expressions of occludin-1 and E-cadherin in the lung tissues significantly increased in the GBFXD group(P<0.05).Compared with those in the FAR group,the expressions of caspase-1,NLRP3,NF-κB and IL-1βin the lung tissues significantly decreased in the GBFXD group(P<0.05).Conclusion:GBFXD may protect the integrity of airway epithelial barrier by regulating the pathway of NLRP3 inflammasome,and improve the respiratory inflammation responses of asthma accompanied with food allergy.
