摘要
目的优化液相色谱仪器条件,建立测定复合消毒液中的葡萄糖酸氯己定含量的方法。方法体积分数为80%的酒精为提取溶剂,在50℃水浴加热条件下,超声波提取消毒剂样品中的葡萄糖酸氯己定。以添加了三氟乙酸的醋酸盐缓冲液-乙腈为流动相,C18色谱柱分离后,258 nm波长处测定葡萄糖酸氯己定含量。结果在5~500 mg·L^(-1)范围内,复合消毒液中葡萄糖酸氯己定含量与色谱峰面积成正比,方法检出限为0.5 mg·L^(-1)。样品加标回收率92.5%~101.3%,相对标准偏差小于4.2%(n=7)。结论高效液相色谱法具有快速简单、专属性强、准确度和精密度好的优点,适用于复配消毒剂中葡萄糖酸氯己定含量的检测。
Objective To optimize the conditions of liquid chromatography instrument,and establish a method for the determination of chlorhexidine gluconate in compound disinfectant.Methods Alcohol with a volume fraction of 80% was the extraction solvent,chlorhexidine gluconate was extracted from disinfectant samples under the condition of 50℃ water bath heating.The content of chlorhexidine gluconate was determined at 258 nm after separation on C18 column with acetonitrile acetate buffer added with trifluoroacetic acid as mobile phase.Results In the range of 5-500 mg·L^(-1),the content of chlorhexidine gluconate was proportional to the chromatographic peak area,and the detection limit was 0.5 mg·L^(-1).The recoveries were 92.5%-101.3% with relative standard deviation less than 4.2%(n=7).Conclusion The method was rapid,simple,specific,accurate and precise.It was suitable for the determination of chlorhexidine gluconate in compound disinfectants.
