摘要
液相色谱-质谱(LC-MS/MS)技术作为一种新型的小分子定性定量的检测手段,在十多年的发展过程中,逐渐成为临床诊断治疗领域最为前沿的技术手段。LC-MS/MS是一种联合液相色谱仪和质谱分析的检测技术,结合了色谱技术高分离能力和质谱对物质结构和质量的分析能力,具有高选择性,高特异性以及高灵敏度的特点。与免疫方法相比,LC-MS更适用于小分子化合物(激素、药物代谢及其产物、维生素、氨基酸)的定性和定量检测。LC-MS在临床的广泛应用,要求实验室的技术人员除具备全方面的专业知识和分析化学等基础学科的掌握,还要进行实验操作和临床报告解读的考核。目前对临床质谱实验人才的培养尚欠缺规范和明确的定义。本文依据已发表的临床质谱实验室应用专家共识,结合本科室质谱团队建设、发展经验,对临床质谱技术人才的培养提出指导意见。
In the past few decades,liquid chromatography-tandem mass spectrometry(LC-MS/MS)technology,as a new qualitative and quantitative detection method,has gradually become the most recognized technology in the field of clinical diagnosis and treatment.LC-MS/MS is an analytical technology combining liquid chromatography and mass spectrometry,which shows a highly efficient molecular separation ability of chromatography technology and the analysis ability of mass spectrometry with ultra-high sensitivity and specificity.Compared with traditional immunoassay,LC-MS/MS is more suitable for qualitative and quantitative detection of small molecules,such as steroid hormones,drug metabolites,vitamins and amino acids.With the wide clinical applications of LC-MS/MS,laboratory technicians are required not only to have theoretical knowledge of mass spectrometry,analytical chemistry and basic statistics,but also to be able to perform the MS-based routine tests for the patients and clinical report interpretation.At present,there is no established standard and clear definition for the training process of laboratory technicians in mass spectrometry application.In this paper,based on the published consensus of clinical mass spectrometry laboratory application,combined with the experience of our own MS laboratory development,suggestions were put forward for the training of clinical mass spectrometry technicians.
