摘要 目的初步探究长链非编码RNA(LncRNA)NEAT1在心力衰竭中的临床意义及作用机制。方法选取2019年8月—2020年10月收治的高血压合并心力衰竭病人与高血压病人为研究对象,检测两组NEAT1与miR-342-3p的表达情况,分析两者对心力衰竭发生、预后复发的预测价值。另取人心肌细胞AC16,应用CCK-8、流式细胞计数检测NEAT1与miR-342-3p对AC16增殖、凋亡能力的影响。结果心力衰竭病人NEAT1呈高表达,在治疗后降低;而miR-342-3p呈低表达,治疗后升高(P<0.05)。受试者工作特征(ROC)曲线分析结果显示,NEAT1与miR-342-3p预测心力衰竭的发生、预后复发均有较好的效果。体外实验表明,沉默NEAT1与过表达miR-342-3p可以使AC16增殖能力增强,凋亡降低;而升高NEAT1与抑制miR-342-3p则反之(P<0.05)。双荧光素酶报告实验显示,NEAT1-WT的荧光活性在转染了miR-342-3p-mi后降低(P<0.05)。且NEAT1-sh组中miR-342-3p表达水平低于NEAT1-sh-NC组,而NEAT1-si组则高于NEAT1-si-NC组(P<0.05)。结论NEAT1在心力衰竭中呈高表达,通过靶向抑制miR-342-3p促进心肌细胞的凋亡参与心力衰竭的发生、发展;两者对于未来临床诊治心力衰竭有着重要的意义。 Objective To explore the clinical significance of long non-coding RNA(LncRNA)NEAT1 in heart failure.Methods Patients with hypertensive heart failure and hypertension were selected as the research subjects.The expression of NEAT1 and miR-342-3p in the two groups were detected,and the prognostic value of the two on the occurrence and development of heart failure was analyzed.In addition,human cardiomyocytes AC16,CCK-8,and flow cytometry were purchased to detect the effects of NEAT1 and miR-342-3p on the proliferation and apoptosis of AC16.Results The expression of NEAT1 in patients with heart failure was high and decreased after treatment,while the expression of miR-342-3p was low and increased after treatment(P<0.05).Receiver operator characteristic(ROC)curve analysis showed that NEAT1 and miR-342-3p could predict the occurrence of heart failure and prognostic recurrence(P<0.05).In vitro experiments showed that silencing NEAT1 and overexpression of miR-342-3p could increase the proliferation of AC16 and reduce apoptosis,while increasing that in NEAT1 and inhibiting miR-342-3p were the opposite(P<0.05).The dual luciferase reporter experiment showed that the fluorescence activity of NEAT1-WT decreased after miR-342-3p-mi was transfected(P<0.05).And the expression level of miR-342-3p in NEAT1-sh group was lower than that in NEAT1-sh-NC group,while that in NEAT1-si group was higher than that in NEAT1-si-NC group(P<0.05).Conclusion NEAT1 is highly expressed in heart failure.By targeting miR-342-3p to promote the apoptosis of cardiomyocytes,it participates in the occurrence and development of heart failure,the results suggest that it is important for the future clinical diagnosis and treatment of heart failure.
