单煎合并与合煎对黄芪-葛根药对配伍广陈皮化学成分影响的指纹图谱研究 被引量:2

摘要 目的采用指纹图谱结合化学计量法以及单煎合并与合煎液研究黄芪-葛根药对配伍广陈皮化学成分的变化。探讨黄芪-葛根药对配伍广陈皮起效的科学内涵。方法以相同工艺参数分别制备黄芪-葛根药对配伍广陈皮单煎合并液与合煎液。基于HPLC指纹图谱,对两者特征峰面积进行主成分分析,确定变量投影重要度(VIP)>1的差异变量(特征峰),采用HPLC进行含量测定。结果两者HPLC指纹图谱特征峰个数一致,OPLS-DA分析结果显示:VIP>1差异变量有9个,VIP>1.25差异变量有3个,由大到小排序:橙皮苷>川陈皮素>桔皮素,从含量测定结果显示,合煎液有效成分含量高于单煎合并液。结论黄芪-葛根配伍广陈皮单煎合并液与合煎液的化学成分种类差异不大,合煎液有利于广陈皮黄酮类成分的溶出,提示黄芪-葛根药对配伍广陈皮降脂协同增效作用可能与橙皮苷、桔皮素和川陈皮素有效成分增加有关。 Objective Fingerprinting combined with stoichiometry,single frying mixed decoction and mixed decoction were used to study the changes of Astragali-Puerariae radix herb pair on the chemical constituents of compatibility with Citri reticulatae Chachiensis pericarpium.Discuss the scientific connotation of Astragali-Puerariae radix herb pair medicine on the effect of compatibility with Citri reticulatae Chachiensis pericarpium.Methods The single frying mixed decoction and mixed decoction of Astragali-Puerariae radix herb pair and Citri reticulatae Chachiensis pericarpium were prepared with the same technological parameters.The fingerprints of the single frying mixed decoction and mixed decoction of Astragali-Puerariae radix herb pair and Citri reticulatae Chachiensis pericarpium were detected.Principal component analysis(PCA)was carried out to determine the difference variable(characteristic peeks)with variable importance for the projection(VIP)>1,subsequently the contents were determined by HPLC.Results The number of characteristic peaks of the two HPLC fingerprints was consistent.OPLS-DA analysis results of the single frying mixed decoction and mixed decoction of Astragali-Puerariae radix herb pair and Citri reticulatae Chachiensis pericarpium show that there were 9 differential variables of VIP>1 and 3 differential variables of VIP>1.25,which were ranked from most to least:Hesperidin,Nobiletin and Tangeretin.Content determination results showed that the content of effective components in mixed decoction were higher than single frying mixed decoction.Conclusion There was little difference in the chemical composition between the single frying mixed decoction and mixed decoction of Astragali-Puerariae radix herb pair and Citri reticulatae Chachiensis pericarpium.Mixed decoction was beneficial to the dissolution of flavonoids from Citri reticulatae Chachiensis pericarpium.It is suggested that the synergistic effect of Astragali-Puerariae radix herb pair on the lipid-lowering effect of Citri reticulatae Chachiensis pericarpium compatibility may be related to the increase of Hesperidin,Nobiletin and Tangeretin.
