基于16S rDNA的HIV/AIDS免疫重建不良患者肠道菌群研究 被引量:4

Research on intestinal microbiota in HIV/AIDS patients with poor immune reconstitution by 16S rDNA

摘要 目的探讨HIV/AIDS患者免疫重建不良、免疫重建良好和健康人群肠道菌群结构、多样性及丰度的差异。方法收集20例免疫重建不良HIV/AIDS患者、20例免疫重建良好HIV/AIDS患者及20例健康人群的粪便样本,采用16S rDNA进行基因测序并进行生物信息学分析。结果HIV/AIDS患者免疫重建不良、免疫重建良好及健康人群的β多样性差异有统计学意义(P<0.05)。菌群构成分析显示,变形菌门、粪杆菌属、大肠埃希菌-志贺菌属、Lachnoclostridium菌、Muribaculaceae菌在三组间丰度差异有统计学意义(P<0.05),其中免疫重建不良组以变形菌门、γ变形菌纲、肠杆菌目、肠杆菌科、大肠埃希菌-志贺菌属、Lachnoclostridium菌、Muribaculaceae菌的菌种相对丰度偏高;免疫重建良好组中瘤胃球菌属的相对丰度较高;健康组中粪杆菌属的相对丰度较高。结论免疫重建不良HIV/AIDS患者、免疫重建良好HIV/AIDS患者和健康人群肠道菌群结构、多样性及丰度存在差异,免疫重建不良HIV/AIDS患者的肠道菌群失调现象更为常见。 Objective To explore the differences in intestinal microflora structure,diversity,and abundance among HIV/AIDS patients with poor immune reconstitution,good immune reconstitution and healthy population.Methods Fecal samples were collected from 20 HIV/AIDS patients with poor immune reconstitution,20 HIV/AIDS patients with good immune reconstitution,and 20 healthy populations.Gene sequencing was performed using 16S rDNA,and bioinformatics analysis was performed.Results There were statistical differences in β diversity among HIV/AIDS patients with poor immune reconstitution,good immune reconstitution,and healthy population(P<0.05).The analysis of bacterial community composition showed that Proteobacteria,Faecalibacterium,Escherichia-Shigella,Lachnoclostridium,and Muribaculaceae had statistical differences in abundance among the three groups(P<0.05).The relative abundance of Proteobacteria,Gammaproteobacteria,Enterobacterales,Enterobacteriaceae and EscherichiaShigella,Lachnoclostridium and Muribaculaceae were higher in poor immune reconstitution group.The relative abundance of Ruminococcus was higher in good immune reconstitution group.The relative abundance of Faecalibacterium was higher in healthy population.Conclusions There were statistical differences in the structure,diversity,and abundance of intestinal microflora in HIV/AIDS patients with poor immune reconstitution,good immune reconstitution and healthy population.The disorder of intestinal microflora appeared ordinary in patients with poor immune reconstitution.
