摘要
目的研究单核细胞趋化蛋白1(MCP-1)和核因子-κB(NF-κB)在右美托咪定注射下的兔脊柱结核免疫应答中的作用。方法新西兰大白兔分成对照组、模型组(脊柱结核模型)、实验低剂量组(脊柱结核模型,2.5μg·kg^(-1)·h^(-1)右美托咪定治疗)、实验中剂量组(脊柱结核模型,5μg·kg^(-1)·h^(-1)右美托咪定治疗)、实验高剂量组(脊柱结核模型,10μg·kg^(-1)·h^(-1)右美托咪定治疗)、阳性组(脊柱结核模型,链霉素、利福平、异烟肼治疗)。用自动血沉分析仪检测红细胞沉降率,用流式细胞仪检测T淋巴细胞亚群,用酶联免疫吸附法检血清中C反应蛋白、MCP-1、肿瘤坏死因子-α(TNF-α)水平,用蛋白质印迹法检测病灶脊柱椎体中NF-κB p65、MCP-1蛋白表达。结果对照组、模型组、实验低剂量组、实验中剂量组、实验高剂量组、阳性组C反应蛋白分别为(14.83±1.33),(135.10±11.27),(91.01±7.71),(70.47±7.67),(39.58±4.07)和(66.19±4.95)μg·L^(-1),红细胞沉降率分别为(2.57±0.22),(13.17±1.64),(9.87±1.33),(7.50±0.49),(5.32±0.46)和(6.32±0.88)mm·h^(-1),CD4^(+)/CD8^(+)分别为1.84±0.16,0.45±0.05,0.69±0.05,0.90±0.08,1.33±0.12和0.97±0.10,MCP-1水平为(195.84±11.79),(337.60±18.17),(292.10±14.62),(247.73±7.29),(206.59±13.36)和(239.75±17.85)pg·mL^(-1),TNF-α水平为(136.13±10.94),(308.22±16.05),(265.69±17.06),(204.63±14.83)和(170.48±14.98),(211.23±17.97)pg·L^(-1)。上述指标,模型组与对照组比较,差异均有统计学意义(均P<0.05);实验低剂量组、实验中剂量组、实验高剂量组、阳性组与模型组比较,差异均有统计学意义(均P<0.05);实验中剂量组与实验低剂量比较,差异均有统计学意义(均P<0.05);实验高剂量组与实验中剂量比较,差异均有统计学意义(均P<0.05)。结论MCP-1和NF-κB可能参与右美托咪定注射下的兔脊柱结核免疫应答调控作用。
Objective To study the immune response of monocyte chemotactic protein-1(MCP-1)and nuclear factor-κB(NF-κB)in rabbit spinal tuberculosis under dexmedetomidine injection.Methods New Zealand white rabbits were divided into control group,model group(spine tuberculosis model),experimental low dose group(spine tuberculosis model,2.5μg·kg^(-1)·h^(-1) dexmedetomidine treatment),experimental medium dose group(spine tuberculosis model,5μg·kg^(-1)·h^(-1) dexmedetomidine treatment),experimental high dose group(spine tuberculosis model,10μg·kg^(-1)·h^(-1) dexmedetomidine treatment),positive group(spine tuberculosis model,streptomycin,rifampicin,isoniazid treatment).Automatic erythrocyte sedimentation rate analyzer was used to detect erythrocyte sedimentation rate,flow cytometry was used to detect T lymphocyte subsets;enzyme linked immunosorbent assay(ELISA)method was used to detect serum C-reactive protein,MCP-1,tumor necrosis factor-α(TNF-α),and Western blot method was used to detect the expression of NF-κBp65 and MCP-1 protein in the vertebral body of the lesion.Results The C-reactive protein of the control group,model group,experimental low dose group,experimental medium dose group,experimental high dose group,and positive group were(14.83±1.33),(135.10±11.27),(91.01±7.71),(70.47±7.67),(39.58±4.07),(66.19±4.95)μg·L-1;erythrocyte sedimentation rate were(2.57±0.22),(13.17±1.64),(9.87±1.33),(7.50±0.49),(5.32±0.46),(6.32±0.88)mm·h^(-1);CD4+/CD8+were 1.84±0.16,0.45±0.05,0.69±0.05,0.90±0.08,1.33±0.12,0.97±0.10;MCP-1 levels were(195.84±11.79),(337.60±18.17),(292.10±14.62),(247.73±7.29),(206.59±13.36),(239.75±17.85)pg·mL-1;TNF-αlevel were(136.13±10.94),(308.22±16.05),(265.69±17.06),(204.63±14.83),(170.48±14.98),(211.23±17.97)pg·L-1.The above indicators,compared with model group and control group,the difference were statistically significant(P<0.05);compared between the experimental low dose group,the experimental medium dose group,the experimental high dose group,the positive group and the model group,the difference were all statistically significant(all P<0.05);compared with the the experimental medium dose group and the experimental low dose group,the difference were all statistically significant(all P<0.05);compared with the experimental high dose group and the experimental medium dose group,the difference were all statistically significant(all P<0.05).Conclusion MCP-1 and NF-κB participate in the regulation of the immune response of rabbit spinal tuberculosis under dexmedetomidine injection.
