摘要
目的探讨microRNA-335-5p对人软骨细胞中WISP3的调节作用,研究microRNA-335-5p在骨性关节炎(OA)中的发病机制。方法通过分离并提取小鼠软骨细胞,分别转染microRNA-335-5p mimics、mimics NC、inhibitor、inhibitor NC质粒,通过qRT-PCR及Western Blotting检测各组中WISP3的表达水平。结果与阴性对照组相比,microRNA-335-5p过表达下调了WISP3的mRNA和蛋白水平的表达,相反microRNA-335-5p低表达上调WISP3的mRNA和蛋白水平的表达。结论microRNA-335-5p能抑制WISP3的mRNA转录及蛋白合成,可能通过靶向软骨细胞中的WISP3对OA的发生及发展起到潜在的作用。
Objective To investigate the regulatory effect of microRNA-335-5p on human chondrocytes WISP3 and the pathogenesis of miR-335-5p in OA.Methods Removal of articular cartilage from traumatic patients,separating and extracting chondrocytes,the expression level of WISP3 in each group was detected by qRT-PCR and Western Blotting after transfection of microRNA-335-5p mimics,mimics NC,inhibitor,inhibitor NC.Results Compared with the negative control group,microRNA-335-5p overexpression downregulated mRNA and protein expression levels of WISP3,on the contrary,the low expression of microRNA-335-5p up-regulated the expression of mRNA and protein in WISP3.Conclusion MicroRNA-335-5p can inhibit mRNA transcription and protein synthesis of WISP3,microRNA-335-5p plays a potential role in the pathogenesis and development of OA by regulating WISP3 in chondrocytes.
