摘要
为分析造纸法再造烟叶生产过程微生物的多样性及其分布与变化情况,以再造烟叶生产过程各物料为研究对象,采用Illumina高通量测序技术对25个物料样品的细菌16S rRNA V3-V4区和真菌ITS序列进行测序分析,获得了生产过程物料细菌和真菌丰度及群落组成情况。结果表明:①再造烟叶生产过程物料中共检测出393个细菌属和165个真菌属,其中优势菌属为乳杆菌属(Lactobacillus)、芽孢杆菌属(Bacillus)、假丝酵母属(Candida)和曲霉属(Aspergillus);②生产过程不同物料微生物群落多样性和结构存在明显差异,聚类分析显示细菌属多样性和结构变化相对较小而真菌属变化相对较大;③从萃取到制涂布液过程中,细菌乳杆菌属(Lactobacillus)相对丰度逐渐增加,而真菌假丝酵母属(Candida)相对丰度总体逐渐降低。
To investigate the diversity and distribution of microorganisms in reconstituted tobacco production process,25 material samples were collected from a reconstituted tobacco production line,and the V3-V4 region of bacterial 16S rRNA and the fungal ITS region were sequenced by Illumina high-throughput sequencing technology.The results showed that:1)A total of 393 bacterial genera and 165 fungal genera were identified,and among them the dominant genera were Lactobacillus,Bacillus,Candida and Aspergillus.2)Significant differences were observed in the diversity and structure of bacterial and fungal communities among these material samples,and cluster analysis suggested that the differences of bacterial community diversity and structure among the bacterial genera were relatively small while those among the fungal genera were relatively large.3)In the processes from extraction to coating solution preparation,the relative abundance of Lactobacillus increased and that of Candida decreased gradually.
