茶树油提取物对Lewis肺癌细胞活性及其移植瘤生长的影响 被引量:1

摘要 茶树油主要由萜类物质组成,具有抗菌消炎、抗病毒等作用。松油烯-4-醇是茶树油的主要成分之一,不同提取工艺下茶树油中松油烯-4-醇与异松油烯所占比例相差较大。利用Lewis肺癌(lewis lung cancer,LLC)细胞及其小鼠移植瘤模型,评价6种茶树油提取物的抗肿瘤作用,为茶树油提取物在肺癌治疗中的应用提供理论依据。用不同浓度(0.1%、0.05%、0.01%)的茶树油提取物处理LLC细胞24 h,MTT法检测细胞活性;建立C57BL/6小鼠LLC移植瘤模型,茶树油提取物瘤内或腹腔注射,每天1次,连续5 d;另设阴性和环磷酰胺阳性对照组。通过测量肿瘤体积计算抑瘤率。结果表明:6种茶树油提取物均能显著降低LLC细胞活性,抑制顺序由大到小为提取物2、5、6、4、1、3。6种茶树油提取物均能抑制小鼠LLC移植瘤生长。瘤内注射时肿瘤抑制作用由大到小为提取物4、2、6、1、5、3,腹腔注射时抑制作用由大到小为提取物4、2、6、3、5、1。综合而言,6种茶树油提取物均能不同程度地抑制小鼠LLC细胞及其移植瘤的生长,松油烯-4-醇或异松油烯为主的茶树油均可抑制LLC细胞及其移植瘤的生长。 Tea tree oil(TTO),mainly composed of terpenoids,has the potential appeared antibacterial,anti-inflammatory and antiviral effects.More information was focused on the terpinen-4-ol in TTO up to now.There was more different percentage of terpinen-4-ol and isoterpinene in TTO at different extract technology.Six TTO extract products were evaluated for the anti-tumor effects by the Lewis lung cancer(LLC)cells and their transplanted tumor models,which will be beneficial for the use of the TTO products in the treatment of lung cancer.LLC cells were treated with different concentrations of TTO extracts named 1 to 6 at 0.1%,0.05%,0.01%concentration were treated LLC cells for 24 h,and the cell viability was detected by MTT assay.The transplanted tumor model in C57 BL/6 mouse was established by injection the LLC cells.The mice bearing LLC tumor were randomly divided into PBS negative control,cyclophospamide positive control and TTO extract tested groups.The mice were injected intratumorally or intraperitoneally with PBS,cyclophosphamide or TTO extracts once a day for 5 consecutive days.Tumor volume were measured and then the rate of tumor inhibitory were calculated.Results showed all of six TTO extracts could significantly reduce the LLC cell viability(P<0.05),the order of the inhibition was:extract 2>extract 5>extract 6>extract 4>extract 1>extract 3.All six TTO extracts inhibited the growth of mouse LLC cell transplanted tumor in time-dependent manner(P<0.05).The order of the inhibitory effect for intratumoral injection was as follows:Extract 4>Extract 2>Extract 6>Extract 1>Extract 5>Extract 3;while the order for intraperitoneal injection was:Extract 4>Extract 2>Extract 6>Extract 3>Extract 5>Extract 1.TTO tested product can inhibit the growth of LLC cells and its transplanted tumors in mice at different degrees.TTO extract containing terpinen-4-ol or isoterpinene mainly displayed inhibition to LLC and its transplanted tumor.
