太子参水提物对亚急性镉中毒小鼠肝损伤的保护作用

Protective effect of aqueous extract of Pseudostellaria Radix on liver injury by subacute cadmium poisoning in mice

摘要 目的 探究太子参(Pseudostellaria Radix,PR)水提物对亚急性镉(Cd)中毒小鼠肝损伤的保护作用。方法 50只昆明种小鼠,雌雄各半,随机分为对照组、模型组和PR高、中、低剂量干预组,每组10只。对照组灌胃生理盐水,其余各组隔天用氯化镉溶液(2.5 mg/kg)灌胃,共4周。造模结束后,对照组、模型组正常饲养;PR高、中、低剂量组每天分别给予PR水提物400、200、100 mg/kg灌胃进行干预,连续20 d后处死各组小鼠,称重法计算肝脏系数,苏木素-伊红(HE)染色观察肝组织病理变化,原子吸收光谱法测定肝组织Cd含量,试剂盒测定小鼠血液白蛋白(albumin,ALB)、酸性磷酸酶(acid phosphatase,ACP)、碱性磷酸酶(alkaline phosphatase,ALP)与肝组织丙二醛(malondialdehyde,MDA)、天冬氨酸氨基转移酶(aspartate aminotransferase,AST)和丙氨酸氨基转移酶(alanine aminotransferase,ALT)水平。结果 与对照组相比,模型组肝脏系数增大(P<0.05),肝组织发生病理变化,肝组织Cd、ALB和MDA含量增加(P<0.05),血清ACP、ALP、ALT、AST活性均升高(P<0.05)。与模型组相比,PR干预组的肝脏系数变小(P<0.05),肝组织Cd、ALB和MDA含量降低(P<0.05),血清ACP、ALP、ALT、AST的活性均下降(P<0.05)。肝组织病理学检查显示,PR高剂量组肝组织病理损伤有明显改善。结论 PR水提物对小鼠亚急性Cd中毒肝损伤具有一定的保护作用。 Objective To explore the protective effect of water extract of Pseudostellaria Radix(PR)on liver injury from subacute toxicity of cadmium(Cd) in mice.Methods Fifty Kunming mice,half male and half male,were randomly divided into control group,poisoning group,and high-dose,medium-dose and low-dose PR interventing groups 10 mice in each group.The control mice were intragastrically administered with normal saline,the other mice were gavaged with cadmium chloride solution(2.5 mg/kg)every other day for four weeks.Then,the mice of control group and poisoning group were fed normally,and the mice of PR interventing groups were given the aqueous extract of PR of 400 mg/kg,200 mg/kg,and 100 mg/kg daily for 20 days.Then,the mice were killed and the liver coefficient was calculated by weighing method;hematoxylin-eosin(HE)staining was used for observing the pathological changes of liver tissue;the content of Cd in liver tissues was determined by atomic absorption spectrometry.Additionally,the related indexes to liver function of mice were detected as well by kits including albumin(ALB)level,the activities of acid phosphatase(ACP),alkaline phosphatase(ALP)serum and the malondialdehyde(MDA)level and the activities of aspartate aminotransferase(AST),alanine aminotransferase(ALT)in liver tissue.Results The results showed that compared with control group,the liver coefficient,liver content of Cd,blood levels of ALB and MDA,and serum activities of ACP,ALP,ALT and AST in mice of poisoning group were strikingly increased(P<0.05)and there were some apparent pathological changes in liver tissue appeared as well;while the liver coefficient,the liver contents of Cd,the blood levels of ALB and MDA and the serum activities of ACP,ALP,ALT and AST in the mice of PR interventing group were all obviously decreased(P<0.05)compared with that of poisoning group,and the pathological examination of liver tissue showed that the abnormal damages in mice of high-dose PR interventing group also had significantly improved.Conclusion The results suggested that the aqueous extract of PR had some protective effect on liver injury from subacute poisoning of cadmium in mice.
