摘要
目的 探究泛素特异性蛋白酶1(USP1)、核转运蛋白α2(KPNA2)在子宫内膜异位症患者在位、异位内膜组织中表达及临床意义。方法 选取海南妇产科医院2019年2月至2021年10月住院的67例子宫内膜异位症患者为研究对象,取其在位内膜组织和异位内膜组织,收集患者剖宫产分娩次数、囊肿直径等临床指标;同期选取行全子宫切除的70例患者的正常子宫内膜组织作为对照。采用实时荧光定量聚合酶链反应(real-time qPCR)法检测组织USP1mRNA、KPNA2 mRNA表达水平,采用免疫组织化学法检测组织USP1、KPNA2蛋白表达水平;采用Pearson法及Spearman等级相关分析异位内膜组织中USP1 mRNA与KPNA2 mRNA及其蛋白表达的相关性。结果 正常子宫内膜组织、在位内膜组织、异位内膜组织中USP1 mRNA、KPNA2 mRNA表达水平及蛋白阳性率依次升高(P<0.05)。异位内膜组织中USP1 m RNA与KPNA2 m RNA表达呈正相关(r=0.646,P<0.05)。异位内膜组织中USP1与KPNA2蛋白表达呈正相关(r=0.589,P<0.05)。子宫内膜异位症患者异位内膜组织中USP1、KPNA2蛋白表达与年龄、剖宫产分娩次数、痛经无关(P>0.05),与囊肿直径、浸润深度有关(P<0.05)。结论 USP1与KPNA2在子宫内膜异位症患者异位内膜组织中呈高表达,且二者与囊肿直径、浸润深度有密切联系。
Objective To explore the expression and clinical significance of ubiquitin-specific protease 1(USP1) and karyopherin α2(KPNA2) in eutopic and ectopic endometrial tissues of patients with endometriosis. Methods Sixty-seven patients with endometriosis who were hospitalized in our hospital during February 2019 to October 2021 were selected as the research objects, the eutopic endometrial tissue and the ectopic endometrial tissue were taken, and the clinical indicators were collected, such as the number of cesarean sections and the diameter of cysts. At the same time, the normal endometrial tissues of 70 patients who underwent total hysterectomy were selected as controls. Real-time quantitative polymerase chain reaction(real-time qPCR) method was used to detect the expression levels of USP1 mRNA and KPNA2 mRNA in tissues,immunohistochemistry was used to detect the expression levels of USP1 and KPNA2 proteins in tissues. Pearson method and Spearman rank correlation were used to analyze the correlation between USP1 mRNA and KPNA2 mRNA and protein expression in ectopic endometrial tissue. Results The expression levels of USP1 mRNA and KPNA2 mRNA and the protein positive rates in normal endometrial tissue, eutopic endometrial tissue, and ectopic endometrial tissue increased sequentially(P<0.05). The expression of USP1 m RNA and KPNA2 m RNA in ectopic endometrial tissue was positively correlated(r=0.646,P<0.05). The protein expression of USP1 and KPNA2 in ectopic endometrial tissue was positively correlated(r=0.589,P<0.05). The expression of USP1 and KPNA2 proteins in the ectopic endometrial tissue of patients with endometriosis had nothing to do with age, number of cesarean delivery and dysmenorrhea(P>0.05), but was related to cyst diameter and depth of infiltration(P<0.05). Conclusion USP1 and KPNA2 are highly expressed in the ectopic endometrial tissue of patients with endometriosis, and the two are closely related to the diameter of the cyst and the depth of infiltration.
