化瘀固本中药方对慢性盆腔炎大鼠盆腔粘连及TGF-β1/Smads信号通路的影响 被引量:4

Effects of Huayu Guben Chinese herbal formula on pelvic adhesion and TGF-β1/Smads signaling pathway in rats with chronic pelvic inflammation

摘要 目的 基于转化生长因子β1(TGF-β1)及信号转导蛋白(Smads)信号通路探讨化瘀固本中药方对慢性盆腔炎(CPID)大鼠盆腔粘连的影响。方法 动物实验分为假手术组、模型组、对照组和实验组,模型组、对照组、实验组SD雌性大鼠采用向卵巢推注0.06 m L的25%苯酚树胶构建慢性盆腔炎大鼠模型,假手术组大鼠推注等剂量的生理盐水。对照组和实验组分别以2 mL剂量的康妇炎胶囊和化瘀固本中药方进行灌胃给药,假手术组和模型组以等剂量的生理盐水进行灌胃处理,每日1次,每组分别连续灌胃2周。处死大鼠剥离子宫。观察大鼠子宫,并对其盆腔粘连进行评分。免疫组化检测子宫组织中细胞间黏附分子-1intercell(ICAM-1)的表达。Western blot检测TGF-β1、p-Smad2/3的表达。结果 与假手术组相比,模型组、对照组、实验组大鼠子宫粘连评分、子宫组织中ICAM-1、TGF-β1、p-Smad2/3的表达明显升高,差异均具有统计意义(P<0.05);与模型组相比,对照组、实验组大鼠子宫粘连评分、子宫组织中ICAM-1、TGF-β1、p-Smad2/3的表达明显降低,差异均具有统计意义(P<0.05)。结论 化瘀固本中药方能够改善慢性盆腔炎大鼠的盆腔粘连症状,其机制可能与抑制TGF-β1/Smads信号有关。 Objective to observe the effect of Huayu Guben Chinese herbal formula on pelvic adhesion in rats with chronic pelvic inflammatory disease(CPID) based on the transforming growth factor-β1(TGF-β1)/Smads signaling pathway.Methods The animal experiment was divided into sham operation group, model group, control group and experimental group.The rats in the model group, control group and experimental group were injected 0.06 mL of 25% phenol gum into the ovary to construct the chronic pelvic inflammatory rat model. The rats in the sham operation group was injected with equal dose of normal saline. The control group and the experimental group were given intragastric administration with Fuyankang capsule and Huayu Guben Chinese herbal formula at a dose of 2 mL, respectively, and the sham operation group and model group were given intragastric treatment with equal dose of normal saline, once a day for 2 weeks. The rat uterus were observed and then the pelvic adhesions were scored. The expression of intercellular-lular adhesion molecule-1(ICAM-1) in uterine tissue was detected by immunohistochemistry. The expression levels of TGF-β1 and p-Smad2/3 were detected by Western blot. Results Compared with the sham group, the uterine adhesion scores and the expression of ICAM-1, TGF-β1, and p-Smad2/3 in the uterine tissue of the model group, the control and experimental group, were significantly increased, and the differences were statistically significant(P<0.05). Compared with the model group, the uterine adhesion scores and the expression of ICAM-1,TGF-β1, and p-Smad2/3 in the uterine tissue of the control and experimental groups were significantly reduced, and the differences were statistically significant(P<0.05). Conclusion Huayu Guben Chinese herbal formula can improve the symptoms of pelvic adhesions in rats with chronic pelvic inflammation, and its mechanism may be related to the inhibition of TGF-β1/Smads signaling.
