摘要
目的:在牙釉质表面制备气相二氧化硅(SiO_(2))/聚二甲基硅氧烷(PDMS)超疏水涂层,并评价涂层的细胞毒性与抗菌性能。方法:将不同质量百分比的亲/疏水SiO_(2)、PDMS溶于正己烷及其他相关助剂中形成悬浮溶液,喷涂于牙釉质表面形成超疏水涂层。采用接触角测量仪测量涂层的静态接触角及动态接触角;扫描电镜(SEM)观察微观结构;X射线光电子能谱(XPS)分析化学组成;傅立叶变换红外吸收光谱仪(FTIR)分析涂层内基团;MTT比色法检测细胞毒性;自动菌落形成实验(CFU)法验证其抗变形链球菌粘附能力。结果:亲/疏水性SiO_(2)质量比为1∶1时,涂层具有最好的疏水效果,其静态接触角(CA)为164°,动态接触角(SA)为1.4°;其超疏水效果来源于纳米SiO_(2)粒子提供的表面粗糙度和PDMS提供的低表面能;MTT结果显示该涂层具有较好的细胞相容性;抗菌实验结果表明,超疏水涂层抗菌效果明显优于对照组,6 h以内抗粘附效果稳定,抗粘附效率达到87%以上。结论:成功在牙釉质表面构建了生物相容性的超疏水涂层,该涂层具有较好的抗变形链球菌粘附能力,为开发口腔防龋喷剂提供思路。
Objective:To prepare fumed silica/polydimethylsiloxane(SiO_(2)/PDMS)superhydrophobic coating on the surface of tooth enamel,and evaluate the cytotoxicity and antibacterial ability of the superhydrophobic coating.Methods:Different mass percentages of hydrophilic and hydrophobic SiO_(2)and PDMS were dissolved in n-hexane and other related additives to form suspension solution and sprayed on enamel surface to form superhydrophobic coating.The contact angle and sliding angle of coating were measured by contact angle measuring instrument.Scanning electron microscopy(SEM)was used to observe the microstructure.X-ray photoelectron spectroscopy(XPS)was used to analyze its element concentration.Fourier transform infrared absorption spectrometer(FTIR)was used to analyze the inner group of the coating.Cytotoxicity test was performed to detect its biocompatibility.Automatic colony forming unit(CFU)was used to verify its anti-streptococcus mutans adhesion ability.Results:The best hydrophobic effect was when the hydrophilic fumed silica:hydrophobic fumed silica was 1∶1.At this moment,the average contact angle reached 164°,and the sliding angle reached 1.4°.The superhydrophobic effect comes from the surface roughness provided by nano-SiO_(2)particles and the low surface energy provided by PDMS.MTT results showed that the coating had good biocompatibility.Antibacterial test showed that the antibacterial effect of superhydrophobic coating was better than that of control group.The anti-adhesion effect was stable within 6 h,reaching more than 87%.Conclusion:A biocompatibility superhydrophobic coating was successfully constructed on the enamel surface.The coating has good adhesion resistance to Streptococcus mutans,which provides ideas for the development of oral caries prevention sprays.
