摘要
目的探讨环状RNA(circRNA)标记物诊断脊柱结核的潜在价值。方法选取2016年6月—2018年9月深圳市第三人民医院收治的50例脊柱结核患者作为研究组,另选择同期进行体检的健康人员50名作为对照组,提取受试者血清RNA实施circRNA基因芯片筛查,将存在差异表达circRNA筛选出后通过实时荧光定量聚合酶链式反应(polymerase chain reaction,PCR)法加以验证。结果circRNA基因芯片初筛获取存在表达差异circRNA36个,表达增高及降低的比例为15/21。经实时定量PCR验证后发现,hsa-circRNA_400005表达量较对照组明显升高,差异有统计学意义(P<0.05),与芯片检测结果基本一致。联合分析circRNA及miRNA表达谱发现,hsa-circRNA_400005与hsa-miR-19a-3p之间存在负相关关系(P<0.05)。结论脊柱结核患者存在大量特异循环表达的circRNA及miRNA,但circRNA的稳定性更高,可作为早期诊断脊柱结核更具潜力的分子标记物。
Objective To investigate the potential value of circular RNA(circRNA)markers in the diagnosis of spinal tuberculosis.Methods 50 patients with spinal tuberculosis who were admitted to Shenzhen Third People's Hospital from June 2016 to September 2018 were selected as the research group,and 50 healthy people who underwent physical examination during the same period were selected as the control group.The serum RNA of the subjects was extracted for circRNA gene chip screening,and the differentially expressed circRNAs were screened out and verified by real-time fluorescence quantitative polymerase chain reaction(PCR).Results 36 circRNAs with differential expression were obtained from the preliminary screening of circRNA gene chip,and the ratio of increased and decreased expression was 15/21.After verification by real-time quantitative PCR,it was found that the expression of hsa-circRNA_400005 was significantly higher than that of the control group,the difference was statistically significant(P<0.05),which was basically consistent with the chip detection results.Combined analysis of circRNA and miRNA expression profiles found that there was a negative correlation between hsa-circRNA_400005 and hsa-miR-19a-3p(P<0.05).Conclusion There are a large number of specific circulating circRNAs and miRNAs in patients with spinal tuberculosis,but circRNAs have higher stability and can be used as molecular markers for early diagnosis of spinal tuberculosis.
