摘要
目的:对薏苡仁饮片、水煎液、配方颗粒的HPLC指纹图谱进行研究,比较其相关性和差异性。方法:采用HPLC-ELSD法测定薏苡仁饮片、水煎液、配方颗粒的指纹图谱,建立三者指纹图谱共有模式,并进行相似度分析。采用皮尔逊相关性分析、聚类分析(CA)、主成分分析(PCA)及正交偏最小二乘判别分析(OPLS-DA)评价三者HPLC指纹图谱的相关性。结果:薏苡仁饮片、水煎液、配方颗粒的HPLC指纹图谱中均确定了8个特征峰,并指认了7个峰;13批薏苡仁饮片、水煎液、配方颗粒指纹图谱与相应对照指纹图谱的相似度均大于0.95,三者对照指纹图谱的相似度均大于0.99。皮尔逊相关性分析结果显示,峰6在薏苡仁饮片、水煎液、配方颗粒之间均不存在显著相关关系。CA和PCA结果显示,薏苡仁水煎液与配方颗粒的关系更为紧密,差异较小。OPLS-DA筛选出3种成分是造成不同批次薏苡仁样品指纹图谱差异性的主要标记物。结论:薏苡仁饮片、水煎液、配方颗粒的主要化学成分组成具有一致性,可为薏苡仁配方颗粒生产过程质量控制及成品质量标准制定提供参考。
Objective: To study the fingerprints of pieces, water decoction and formula granules of semon coicis, and compare their correlation and difference. Methods: HPLC-ELSD was adopted to establish the fingerprints of pieces, water decoction and formula granules of semon coicis. The common pattern of the three fingerprints was established and the similarity was analyzed. Pearson correlation analysis, clustering analysis(CA), principal component analysis(PCA) and orthogonal partial least squares discriminant analysis(OPLS-DA) were used to evaluate the correlation of the three HPLC fingerprints. Results: Eight characteristic peaks were determined in the HPLC fingerprints of pieces, water decoction and formula granules of semon coicis, and among them, seven peaks were identified. The similarity among the fingerprints of 13 batches of pieces, water decoction and formula granules of semon coicis and the corresponding control fingerprint was greater than 0.95, and the similarity among the three control fingerprints was greater than 0.99. Pearson correlation analysis showed that there was no significant correlation in peak 6 among the fingerprints of pieces, water decoction and formula granules of semon coicis. CA and PCA results showed that the relationship between water decoction and formula granules of semon coicis was closer, and the difference was small. OPLS-DA screened three components as the main markers that caused the difference in the fingerprints of semon coicis samples with different batches.Conclusion: The main chemical compositions of pieces, water decoction and formula granules of semon coicis are basically the same, which provide reference for the quality control of production process of semon coicis formula granules, and the formulation of quality standard for finished products.
