醒神健脑方对非痴呆性血管性认知障碍大鼠同型半胱氨酸及氧化应激反应的影响 被引量:2

摘要 目的 观察醒神健脑方对非痴呆性血管性认知障碍大鼠同型半胱氨酸(Hcy)及氧化应激反应的影响。方法 将60只SD大鼠随机分成3组,每组20只。空白组给予普通饲料喂养,模型组和醒神健脑方组给予3%蛋氨酸饲料喂养建立高同型半胱氨酸血症非痴呆性血管性认知障碍模型。造模成功后,醒神健脑方组给予醒神健脑方灌胃,空白组和模型组给予等量蒸馏水灌胃,均连续灌胃14 d。灌胃结束后进行水迷宫实验,取血检测Hcy、一氧化氮(NO)、超氧化物歧化酶(SOD)、丙二醛(MDA)水平。结果 定位航行实验中,各组大鼠的逃避潜伏期均逐渐缩短,除第4天外,空白组和醒神健脑方组均明显短于模型组(P均<0.05);空间探索实验中,空白组和醒神健脑方组大鼠进入目标象限次数均明显多于模型组(P均<0.05),目标象限停留时间均明显长于模型组(P均<0.05)。与空白组比较,模型组大鼠血清Hcy、MDA水平均明显升高(P均<0.05),血清SOD、NO水平均明显降低(P均<0.05);与模型组比较,醒神健脑方组大鼠血清Hcy、MDA水平均明显降低(P均<0.05),血清SOD、NO水平均明显升高(P均<0.05)。结论 醒神健脑方可明显改善非痴呆性血管性认知障碍大鼠行为认知功能,机制可能与降低Hcy水平及减轻氧化应激反应有关。 Objective It is to observe the effect of Xingshen Jiannao decoction on homocysteine(Hcy)and oxidative stress in rats with vascular cognitive impairment no dementia(VCIND).Methods Sixty SD rats were randomly divided into 3 groups with 20 rats in each group.The blank group was fed with normal diet,the model group and the Xingshen Jiannao decoction group were fed with 3%methionine diet to establish the models of hyperhomocysteinemia and VCIND.After successful modeling,the Xingshen Jiannao decoction group was given Xingshen Jiannao decoction by gavage,and the blank group and the model group were given the same amount of distilled water by gavage,all the groups were treated for 14 days.At the end of treatment,water maze test was performed in the rats,and their blood was collected to detect the contents of Hcy,nitric oxide(NO),superoxide dismutase(SOD)and malondialdehyde(MDA).Results In the positioning navigation experiment,the escape latency of the rats in each group was gradually shortened.Except for that on the fourth day,the latency of the blank group and the Xinshen Jiannao decoction group were significantly shorter than that of the model group(all P<0.05);in the space exploration experiment,the times of entering the target quadrant of the blank group and Hexingshen Jiannao decoction group were significantly more than those of the model group(both P<0.05),and the duration of stay in the target quadrant was significantly longer than that of the model group(P<0.05).Compared with the blank group,the levels of serum Hcy and MDA of the model group were significantly increased(both P<0.05),and the levels of serum SOD and NO were significantly decreased(both P<0.05).compared with the model group,the levels of serum Hcy and MDA of the rats in the Hexingshen Jiannao decoction group were significantly decreased(both P<0.05),and the levels of serum SOD and NO were significantly increased(both P<0.05).Conclusion Xingshen Jiannao decoction can obviously improve the behavior and cognitive function of rats with VCIND,and the mechanism may be related to down-regulating the level of Hcy and relieving oxidative stress.
