Influence of Ultrafine Comminution Technology on Gardenia

摘要 目的:比较超微粉碎前后栀子的变化,并对超微粉碎后栀子的质量进行评价,以考察栀子经超微粉碎后的效果。方法:分别通过显微法、薄层色谱法、高效液相色谱法三种方式对超微粉碎前后的栀子进行质量评价。其中薄层色谱法使用栀子对照药材和栀子苷对照品进行比较,高效液相色谱法中以栀子苷为含量检测指标。结果:粗粉在显微镜下能观察到完整的细胞组织,能进行栀子的显微鉴别,而超微粉在显微镜下已不能见到完整的组织细胞,不具有显微鉴别特征;粗粉和超微粉在薄层色谱法显示相同的效果;高效液相色谱法中超微粉碎不仅没有改变栀子中的功能性成分,还提高栀子苷的提取率,超微粉比粗粉平均提高了17.2%。结论:通过薄层色谱法和高效液相色谱法鉴别发现,超微粉碎技术对栀子中药物活性成分的提取率有大幅度的提升。 Objective:To compare the ultrastructural changes of gardenia before and after the superfine grinding,evaluate the quality of gardenia after superfine comminution,and study the effect of gardenia by superfine grinding.Methods:Three methods are used to evaluate the quality of gardenia after superfine comminution,including microscopy,thin layer chromatography,and high performance liquid chromatography.The thin layer chromatography compares gardenia control medicine with geniposide,and the high performance liquid chromatographic method takes geniposide as content detection index.Results:the coarse powder can be observed in intact cells under a microscope,and the microscopic identification of gardenia can be made;regarding superfine powder,the complete tissue cells can not be seen under the microscope,which does not have the microscopic characteristics;coarse and ultrafine powder shows the same effect by using the thin layer chromatography;when applying the high performance liquid chromatography,superfine grinding does not change the functional components of gardenia,but increases the extraction rate of geniposide.Ultrafine powder increases averagely by 17.2% compared with coarse powder.Conclusion:By using the thin layer chromatography and high performance liquid chromatography identification,superfine comminution technology has greatly improved the extraction rate of active pharmaceutical ingredients of gardenia.
