摘要
探讨长期给SD大鼠灌胃纳米氧化铈后,对SD大鼠的整体健康状况影响。将40只SPF级健康雄性SD大鼠随机分为4组:对照组、低剂量组、中剂量组、高剂量组;每组10只大鼠。实验组分别给予20 mg/kg、100 mg/kg、500 mg/kg纳米氧化铈溶液,对照组给予同等体积去离子水,隔天给大鼠灌胃,实验过程共90天。实验结束后检测大鼠体重、脏器系数、血液生化指标和组织病理学改变。大鼠的体重、脏器重量、脏器系数以及血液生化指标均未发现有实际意义的改变;组织病理学结果发现,肝脏和肾脏组织未见明显改变;在各实验组,空肠组织粘膜上皮有炎症细胞浸润,但未导致空肠炎症发生;对照组和低剂量组的回肠和盲肠组织未见明显损伤,中剂量组和高剂量组在粘膜层均发现肠绒毛结构和腺体受损,并可见大量炎症细胞浸润。长期给大鼠灌入纳米氧化铈,对大鼠的整体毒性较小,轻微损伤多集中于回肠和盲肠。
The effect of long-term intragastric administration of nano-cerium oxide to SD rats on the overall health of SD rats was investigated.Forty SPF-grade healthy male SD rats were randomly divided into 4 groups of 10 rats each:control group,low-dose group,middle-dose group,and high-dose group.The experimental group was given 20 mg/kg,100 mg/kg,500 mg/kg nano cerium oxide solution,and the control group was given the same volume of deionized water,and the rats were given intragastric gavage every other day.The experiment process was 90 days in total.After the experiment,the rats’body weight,organ coefficient,blood biochemical indexes and histopathological changes were detected.Results showed that significant changes were found in the body weight,organ weight,organ coefficient and blood biochemical indexes of the rats;the histopathological results showed that the liver and kidney tissues were not significantly changed;in each experimental group,there was inflammatory cell infiltration in the epithelium of the jejunum tissue mucosa,but it did not cause inflammation of the jejunum;there was no obvious damage to the ileum and cecum tissues of the control group and the low-dose group.In the medium-dose group and the high-dose group,the intestinal villi structure and glands were damaged in the mucosal layer.And a large number of inflammatory cell infiltration can be seen.Long-term infusion of nano-cerium oxide to rats is less toxic to rats as a whole,and the minor damage is mostly concentrated in the ileum and cecum.
