摘要
目的建立黄连-苦参药对不同配伍比例的的指纹图谱,探讨黄连-苦参不同的配伍比例的含量差异。方法采用APS-2 HYPERSIL氨基柱;流动相为乙腈:0.1%磷酸水,梯度洗脱;流速:1.0mL/min,检测波长220 nm,柱温室温;进样量:20 μL,建立黄连-苦参不同比例的指纹图谱,研究有无新成分的产生和主要化学成分含量的动态变化。结果建立了不同比例配伍的黄连-苦参药对的指纹图谱,结果各共有峰的分离度良好,并对共有峰进行了归属。通过HPLC法测得黄连-苦参配伍1:1时主要生物碱含量最高。结论该研究建立的黄连-苦参指纹图谱测定方法,揭示了药对配伍的量效关系,为药对配伍的进一步研究提供了依据。
Objective To establish the finger prints of the couplet medicines of golden thread and lightyellow sophora root in their different proportions,and to explore their content differences with different compatibility ratios.Methods Gradient elution HPLC was used to analyze the content differences of the two medicines on the APS-2 HYPERSIL amino column,the mobile phase:acetonitrile 0.1%phosphoric acid water,with the flow rate at 1.0 mL/min,the detection wavelength at 220 nm,the column temperature:room temperature,and the sample size:20 μ L.The corresponding finger prints of golden thread and lightyellow sophora root in different proportions were established.Whether new components were produced and the dynamic changes of main chemical components were investigated as well.Results The finger prints of golden thread and lightyellow sophora root in different proportions were established.The results showed that the resolution of the common peaks was good,while the common peaks were assigned as well.The contents of main alkaloids in the decoction were the highest when the compatibility ratio of the couplet medicines was 1:1.Conclusion The study establishes the finger prints of golden thread and lightyellow sophora root,reveals the dose-effect relationships of the couplet medicines,which provides a basis for the further research on the compatibility of couplet medicines.
