一种鼠骨性关节炎低温造模装置的设计及使用效果评价

摘要 目的 :针对传统骨性关节炎模型建立时存在的固定动物花费时间长、动物挣扎剧烈、单人操作困难、低温装置控温难等问题,设计一种操作安全且简便的鼠骨性关节炎低温造模装置。方法:该装置由控温箱和鼠固定器组成,主要制作材料为有机玻璃和金属。其中控温箱主要由卡位条和温度显示屏组成,用于安装鼠固定器和显示温度变化;鼠固定器主要由固定底板、防护罩、第一挡板和第二挡板组成。采用该装置对3月龄BALB/c雄性小鼠进行30 d的低温处理,观察小鼠的后肢形态、关节肿胀度、血清学指标和踝关节病理切片等,验证该装置建立小鼠骨性关节炎模型的可行性。结果:采用该装置可在1 min内完成单只小鼠的固定,且5~6℃低温可稳定维持3 h,整个造模过程小鼠挣扎程度小,操作者未出现任何抓伤或咬伤;小鼠后肢形态、关节肿胀度、血清学指标和踝关节病理切片结果表明,该装置可成功建立鼠骨性关节炎模型。结论:该装置操作简便、安全可靠、低温条件稳定、省时省力,可用于建立小鼠骨性关节炎模型。 Objective To design a safe and convenient low-temperature modeling device for murine osteoarthritis to eliminate the deficiencies of traditional models in long time spent for animal immobilization, animal’s violent struggling,difficult single-person operation and temperature control of the low-temperature unit. Methods A low-temperature modeling device was developed for murine osteoarthritis consisting of a temperature control box and a murine holder, mainly made of organic glass and metal. The temperature control box was made up of spacing bars and a temperature display for mounting the murine immobilizer and showing temperature changes;the murine holder was mainly composed of a fixing base plate, a protective cover, a first stopper and a second stopper. Thirty-day low-temperature treatment was carried out for the 3-month-old BALB/c male mice with the device developed. The morphology of the mouse hind limbs, joint swelling, serological indexes and ankle joint pathology sections were observed to verify the feasibility of the device to establish a mouse osteoarthritis model. Results The device developed could complete fixing a single mouse in one minute and maintain 3 h low temperature(5 to 6 ℃), with no mouse’s violent struggling and no any scratch or bite injury of the operator during modeling;the results of the morphology of mouse hind limbs, joint swelling, serological indexes and pathological sections of ankle joint showed that the device could successfully construct a murine osteoarthritis model. Conclusion The device developed gains advantages in easy operation, high safety and reliability, maintaining stable low-temperature conditions and high efficiency when used to create a mouse osteoarthritis model.
