鼻咽癌高癌家族中热休克蛋白70的表达及意义 被引量:2

摘要 目的探讨热休克蛋白HSP70在鼻咽癌高癌家系癌患者中的表达特点及其生物学意义。方法鼻咽癌高癌家族组和散发组鼻咽癌患者各20例,健康对照者20例,分别采集外周静脉血标本,分离外周血单个核细胞(PBMC),常规抽提总蛋白和总mRNA。然后,分别以Western blotting技术检测PBMC的HSP70蛋白表达水平,RT-qPCR检测PBMC的HSP70 mRNA转录水平,比较分析组间差异及其生物学意义。结果鼻咽癌高癌家族组和散发组鼻咽癌患者以及健康对照组PBMC HSP70蛋白水平分别为0.648±0.292,0.490±0.314,0.395±0.165,组间差异统计学意义显著(P=0.007),高癌组明显高于健康对照组,散发组则否。但是,三组的HSP70 mRNA转录水平则分别为-14.071±2.408,-13.952±2.726,-13.905±1.601,组间差异无统计学意义(P=0.970)。结论受家族遗传易感性影响,在鼻咽癌高癌家族鼻咽癌患者中,致癌因子可能通过转录后调控机制上调鼻咽癌患者PBMC HSP70蛋白的表达,促进鼻咽癌高癌家族遗传易感性鼻咽癌的发病。 Objective To investigate the expressive characteristics and biological implications of heat shock protein 70(HSP70)in patients with nasopharyngeal carcinoma(NPC)derived from high-NPC family.Methods Candidates enrolled in this study included 20 NPC cases derived from high-NPC family(HNPCF),20 cases of sporadic NPC(SNPC),and 20 healthy persons(HP),with their peripheral vein blood samples collected and mononuclear cells separated by density gradient centrifugation respectively.Then,Western blotting procedure was used to detect the expressive level of HSP70 protein and RT-qPCR assay used to determine the transcription level of HSP70 mRNA in these peripheral blood mononuclear cells(PBMC)following total protein and mRNA separated respectively for a comparative analysis to explore their biological implications among different groups.Results The expressive levels of HSP70 protein in PBMC of HNPCF,SNPC and HP groups were 0.648±0.292,0.490±0.314,and 0.395±0.165 respectively,with the highest level seen in HNPCF group and the lowest level in HP group,showing very significantly statistical significance(P=0.007).However,the transcriptional levels of HSP70 mRNA in PBMC among these three groups were-14.071±2.408,13.952±2.726,and 13.905±1.601 respectively,with no significantly statistical significance among them(P=0.970).Conclusions Based on this study,it might be concluded as that,under the action of familial genetic susceptibility,some associated carcinogenic factors can up-regulate the expressive level of HSP70 protein in PBMC via post-transcriptional regulation mechanism to promote the developing risk of familial NPC among members in high-NPC family.
