摘要
研究旨在建立一种检测猪用添加剂预混合饲粮中米诺地尔的超高效液相色谱测定方法。样品采用甲醇振荡提取,混合型强阳离子固相萃取小柱(MCX)净化,采用配有二极管阵列紫外检测器的超高效液相色谱仪测定,外标法定量。结果显示:标准品浓度为0.05~10.00 mg/L,米诺地尔具有良好的线性关系,线性相关系数(R^(2))为0.9996。样品中米诺地尔的最低检出限为2 mg/kg,定量限为5 mg/kg。在5~200 mg/kg范围内,平均回收率为73.6%~89.7%,相对标准偏差(RSDs)均小于4.5%。研究表明,试验建立的方法准确性好、灵敏度高,可有效应用于猪用添加剂预混合饲粮中米诺地尔的检测。
The study was to develop a method for the determination of illegal additives minoxidil in additive premixed feed for pigs by ultra performance liquid chromatography(UPLC).The samples were extracted by methanol shaking,purified by a mixed-type strong cation solid phase extraction cartridge(MCX),determined by an ultra-high performance liquid chromatograph equipped with a diode array UV detector,and quantified by external standard method.The results showed that when standard concentration was in 0.05~10.00 mg/L,minoxidil had a good linear relationship,and the linear correlation coefficient(R^(2))was 0.9996.The minimum detection limit of minoxidil in samples was 2 mg/kg,the quantification limit was 5 mg/kg.The average recoveries were 73.6%~89.7%in the range of 5~200 mg/kg,and the relative standard deviations(RSDs)were all less than 4.5%.The study indicates that the method established in the study has good accuracy and high sensitivity,which can be effectively applied to confirmation and quantification of minoxidil in pig additive premix feed.
