Dobner三组分一锅法制备辛可芬

One-pot Synthesis of Cinchophen Via Three-component Dobner Reaction

摘要 以丙酮酸、苯甲醛和苯胺为原料,乙醇为溶剂,通过“三组分一锅法”在温和条件下制备喹啉类化合物辛可芬。在文献基础上对反应条件进行了优化,以适合本科生实验的开设。根据反应体系中各物质pKa和溶解性的差异,经简单萃取操作,即可得到纯度较高的产物,并通过核磁共振波谱及红外光谱对所得产物进行了结构表征。本实验具有原料廉价易得、操作简单、产率高、溶剂体系环境友好等优点,可作为本科三年级综合化学实验进行开展。本实验的开展有助于帮助学生熟悉喹啉环的合成方法,初步掌握化合物分离提纯和结构表征技术,训练学生的科学思维,以及培养学生的科研创新能力。 Cinchophen(2-phenyl-4-quinolinecarboxylic acid)was assembled from pyruvic acid,benzaldehyde and aniline in ethanol through a three-component one-pot synthesis under mild conditions.On the basis of previous reports,the reaction conditions were optimized to facilitate undergraduate students.After completion of the reaction,the product could be easily obtained from the reaction mixture by extraction,which could then be identified by NMR and IR spectroscopy.This reaction possessed a number of benefits,such as low cost,easy operation,high yields,and ecofriendly solvent systems,which could be introduced as a comprehensive experiment for undergraduate students at the junior year.It would be helpful for students to get to know the synthetic methods of quinolines,learn how to separate and characterize compounds,and improve their scientific research capability.
