亚抑菌浓度亚胺培南西司他丁钠诱导铜绿假单胞菌生物被膜形成的体外研究 被引量:2

摘要 目的研究亚抑菌浓度(Sub-MIC)下,亚胺培南西司他丁钠(IM/CI)对铜绿假单胞菌(PA)临床分离株(PA.12)生物被膜形成的影响。方法分别将1/2 MIC、1/4 MIC、1/8 MIC IM/CI作用于PA.12,通过细菌黏附力试验了解Sub-MIC IM/CI对PA.12黏附力的影响。通过运动试验、结晶紫染色及激光共聚焦显微镜(CLSM)观察1/4 MIC IM/CI对PA.12生物被膜形成的影响,RT-PCR检测1/4 MIC IM/CI对PA.12生物被膜形成相关基因algC、algD的转录水平。结果1/4 MIC、1/8 MIC IM/CI能增强PA.12黏附力;1/4 MIC IM/CI能增强PA.12的运动能力,促进生物被膜形成,增加生物被膜厚度,同时能增强PA.12生物被膜形成相关基因algC、algD的转录水平。结论1/4 MIC IM/CI可诱导PA.12生物被膜形成。 Objective To investigate the effect of imipenem cilastatin sodium(IM/CI)on the biofilm formation of pseudomonas aeruginosa(PA.12)under subminimal inhibitory concentration(Sub-MIC).Methods One half MIC,1/4 MIC and 1/8 MIC IM/CI were applied in PA.12,respectively.The effect of IM/CI on the adhesion of PA.12 was investigated by bacterial adhesion test.The influence of the 1/4 MIC IM/CI on PA.12 biofilm formation was observed by exercise test,crystal violet staining and Confocal laser scanning microscope(CLSM).The transcriptional levels of algC and algD genes related to the biofilm formation of PA.12 were detected by RT-PCR.Results Both 1/4 MIC and 1/8 MIC IM/CI could enhance the adhesion of PA.12.Only 1/4 MIC IM/CI enhanced the swimming ability and swarm movement ability of PA.12 to promote the formation of biofilm and increase the thickness of biofilm and upregulate the transcription levels of algC and algD genes related to biofilm formation of PA.12.Conclusion A total of 1/4 MIC IM/CI could induce the formation of PA.12 biofilm.
