摘要
目的:采用平喘方干预哮喘模型观察IL-33-ST2通路在肺肠组织的变化研究。方法:40只BALB/c小鼠分空白对照组、模型组、地塞米松组(DEX)和平喘方组(PCF)。用OVA联合Al(OH)3建造哮喘模型。通过HE染色观察肺和肠道组织形态学变化。ELISA检测BALF中IL-5和IL-13的变化。通过RT-PCR和Western blot检测肺和肠道组织中IL-33-ST2通路上相关因子的mRNA和蛋白的表达。结果:平喘方能够有效下调哮喘小鼠肺部IL-5和IL-13(P<0.05)的表达,其中IL-13的表达差异最为显著。同时能够下调肺肠组织中IL-33-ST2通路上相关因子,其中IL-33等表达差异显著(P<0.05)。结论:平喘方下调哮喘模型肺肠组织IL-33-ST2通路中相关因子的表达。
Objective:Observation on the changes of lung and intestinal immunity of IL-33-ST2 in asthma model intervened by Pingchuan formula.Methods:Forty BALB/c mice were divided into control group,model group,dexamethasone group(DEX)and Pingchuan formula group(PCF).The asthma model was established by OVA combined with Al(OH)3.The morphological changes of lung and intestine were observed by HE staining.The changes of IL-5 and IL-13 in BALF were detected by ELISA.RT-PCR and Western blot were used to detect the related mRNA and protein expression of IL-33-ST2 pathway in lung and intestine.Results:Pingchuan formula could effectively down-regulate the expression of IL-5 and IL-13 in the lungs of asthmatic mice(P<0.05),and the most obvious inhibition was the expression of IL-13.At the same time,it could down regulate the related factors of IL-33-ST2 pathway in lung and intestine tissues,and the differences of IL-33 and other factors were significant(P<0.05).Conclusion:Pingchuan formula can downregulate the expression of IL-33-ST2 pathway related factors in lung and intestine of asthma model.
